目的 观察凤尾草总黄酮对慢性非细菌性前列腺炎模型大鼠的治疗作用,探讨其作用机制.方法 Wistar雄性大鼠58只,腹腔注射大鼠前列腺蛋白提纯液辅以免疫佐剂制备非细菌性前列腺炎大鼠模型,造模后随机分为空白组、模型组、凤尾草低中高剂量组、普适泰组,每组8只,灌胃给予生理盐水及相应药物,连续给药30 d,观察大鼠体质量及前列腺湿重变化及免疫器官重量变化,光镜下观察各组大鼠前列腺组织的炎性细胞浸润,并应用酶联免疫吸附试验(ELISA)法检测大鼠血清中白细胞介素(IL)-8、IL-10、肿瘤坏死因子-α(TNF-α)的水平.结果 凤尾草各组并不引起大鼠的体质量、前列腺湿重的明显变化,凤尾草高剂量组大鼠脾脏胸腺重量[(0.643 ±0.042)、(0.287 ±0.042)g]与模型组[(0.701 ±0.054)、(0.354±0.051)g]比较明显下降,差异有统计学意义(P <0.05);组织病理学观察提示凤尾草总黄酮能明显改善模型大鼠前列腺组织结构破坏及间质水肿,降低炎性细胞浸润水平,其中凤尾草高剂量组改善最为明显.与空白组[(4.69±0.67)、(79.95 ±2.84)、(44.73±1.53) ng/L]比较,模型组血清IL-8、IL-10、TNF-α水平[(5.31±0.26)、(88.94±3.12)、(47.34±1.91) ng/L]均有所升高,差异有统计学意义(P<0.05);凤尾草高剂量组血清IL-8和IL-10水平[(5.07±0.42)、(82.32±3.09) ng/L]比模型组[(5.31±0.26)、(88.94±3.12) ng/L]明显降低,差异有统计学意义(P<0.05),其血清TNF-α水平(44.73±1.53)比(47.34±1.91) ng/L差异有统计学意义(P<0.01);凤尾草中剂量组血清IL-8和TNF-α水平[(5.06±0.30)、(44.80±2.20) ng/L]明显降低,与模型组比较差异有统计学意义(P<0.05).结论 凤尾草总黄酮各剂量组均能改善大鼠慢性非细菌性前列腺炎组织病理学变化,其中凤尾草高剂量组改善明显,其作用机制可能与调节大鼠的免疫功能,平衡炎性因子的产生有关.

Objective To study the therapeutical effects of total flavonoid of pteris multifida in nonbacterial prostatitis rats and the possible mechanism.Methods Fifty-eight male Wistar rats were intraperitoneally injected with the Wistar rat prostate protein purification liquid mixture with immune adjuvant for the preparation of experimental rat model of chronic nonbacterial prostatitis.The rats were randomly divided into 6 groups (8 rats each group):the blank group,model group,total flavonoid of pteris multifida group (high,medium,and low dose groups),prostat group.The rats were fed on relevant drugs or the same volume of distilled water in each group,once daily for consecutive 30 days.The body weight,the immune organs weight and the prostate wet weight were measured,and the prostatic index (PI) was calculated.The levels of interleukin (IL)-8,IL-10 and tumor necrosis factor-α (TNF-α) were determined by enzyme linked immunosorbent assay (ELISA) assay kit.The pathological changes were observed in the prostate tissues under the light microscope.Results Total flavonoid of pteris multifida could not reduce the body weight and prostate wet weight.As compared with model group [(0.701 ±0.054),and (0.354 ±0.051) g],the spleen weight and thymus weight in high dose total flavonoid of pteris multifida treated group [(0.643 ± 0.042),and (0.287 ± 0.042) g] were significantly reduced (P < 0.05).Under the light microscopy,total flavonoid of pteris multifida could obviously alleviate the pathological changes in the prostate tissue in rat models,reduce infiltration of inflammatory cells,most significantly in the high dose total flavonoid of pteris multifida treated group.As compared with blank group [(4.69 ± 0.67),(79.95 ±2.84),and (44.73 ± 1.53) ng/L],IL-8,IL-10 and TNF-α levels were remarkably increased in model group [(5.31 ± 0.26),(88.94 ± 3.12),and (47.34 ± 1.91) ng/L,P < 0.05].As compared with model group,the serum levels of IL-8 and IL-10 in high dose total flavonoid of pteris multifida treated group was remarkably depressed [(5.07 ± 0.42,82.32 ± 3.09) vs.(5.31 ± 0.26,88.94 ±3.12) ng/L,P <0.05].As compared with model group [(47.34 ± 1.91) ng/L],the serum levels of TNF-α in high dose total flavonoid of pteris multifida treated group [(44.73 ± 1.53) ng/L] was significantly reduced (P < 0.01).The levels of IL-8 and TNF-α [(5.06 ± 0.30),(44.80 ±2.20) ng/L] were reduced in the middle dose total flavonoid of pteris multifida treated group group as compared with model group (P < 0.05).Conclusion The total flavonoids of pteris multifida could significantly alleviate the pathological changes of the prostate tissues in rats with chronic nonbacterial prostatitis,most significantly in the high dose total flavonoid of pteris multifida treated group,which might be related to the regulation of the immune function and balance of the production of inflammatory factors.
