目的 建立兔膝关节置换(TKA)术后金黄色葡萄球菌感染模型,检测复合涂层人工关节假体兔体内的抗菌性能.方法 设计制作兔膝人工关节假体,48只新西兰兔随机分为对照组和实验A、B组(n=16).实验A组植入0.5mmol/L纳米银/羟基磷灰石(Ag/HA)人工关节假体;实验B组植入1.0 mmol/L Ag/HA人工关节假体;对照组植入普通人工关节假体.术后14 d膝关节腔内注入金黄色葡萄球菌液0.5 ml(1×106 CFU/ml).检测兔体温、C反应蛋白(CRP)、红细胞沉降率(ESR)、血清降钙素原(PCT)水平的变化.2周后处死兔,膝关节周围组织取材行细菌培养.结果 对照组兔体温、血清CRP值、ESR值、PCT值均高于实验组,组间差异有统计学意义(P<0.05).实验A组感染率为23.08%(3/16);实验B组为12.50%(2/16);对照组为100.00%(16/16).实验组与对照组之间差异有统计学意义(P<0.05);实验A、B组之间差异无统计学意义(P>0.05).结论 Ag/HA复合涂层人工关节假体动物体内对金黄色葡萄球菌有良好的抗菌活性.

Objective To establish Staphylococcus aureus infective rabbits model after knee replacement,detect the antibacterial activity on the compositive coating.Methods 48 New Zealand white rabbits were randomly divided into three groups:the control group、the experiemental group A and B (n =16).In the group A,0.5 Ag/HA artificial joint prosthesis were implanted;in the group B,1.0 Ag/HA artificial joint prosthesis were implanted;in the control group,normal artificial joint prosthesis were implanted.Mter 14 days,0.5 ml(1 × 106 CFU/ml) were injected into the knee joint cavity,and then obseved the level of the temperature,C-reactive protein (CRP),erythrocyte sedimentation rate (ESR),procalcitonin (PCT).killed the rabbits after 2 weeks and selected the knee joint's tissue to bacterial culture.Results The control group' s temperature,CRP,EST,PCT are higher than the experiemental groups,there is a statisitically significant difference between groups(P <0.05).Infection:The group A is 23.08%;the group B is 12.50%;the control group is 100.00%;There was a statistically significant difference between the experimental group and the control group (P < 0.05);there is no statistically significant differences between the experimental A,B group (P > 0.05).Conclusion Ag/HA composite coating of artificial joint prosthesis has a good antibacterial activity against Staphylococcus aureus in animals.
