目的 观察重组人促红细胞生成素(EPO)对大鼠股骨头坏死的预防作用.方法 将雄性SD大鼠60只随机分为3组,分别为空白组、对照组、实验组.将对照组再分为组A和组B:组A每周一、周五在大鼠臀肌处注射醋酸泼尼松龙50 mg/kg,1次/天,持续4周;组B在进行造模之前3h在大鼠臀肌处注射同EPO等体积的0.9%氯化钠注射液,其余处理措施同组A.实验组每次造模前3h在大鼠臀肌处注射EPO溶液,5000 IU/kg.结果 (1)骨陷窝率:苏木素-伊红(HE)染色切片显示实验组空骨陷窝率为(14.70±1.78)%,对照组为(23.21±2.34)%,空白组为(7.45±1.23)%;(2)血液中脂质含量:分别于1、2、4周检测血液中三酰甘油含量结果如下:空白组为(1.25±0.32)、(1.28 ±0.26)、(1.27±0.40) mmo/L;对照组为(2.17±0.23)、(2.48±0.41)、(1.88 ±0.38) mmo/L;实验组为(1.42±0.34)、(1.47±0.26)、(1.50±0.49) mmo/L.总胆固醇含量分别在1、2、4周变化:空白组:(1.62±0.13)、(1.62±0.14)、(1.62±0.19) mmo/L;对照组:(2.41±0.18)、(2.52±0.24)、(2.13±0.37) mmo/L;实验组:(1.64±0.18)、(1.69±0.24)、(1.66±0.31)mmo/L.(3)组织学检测:实验组与对照组比较,股骨头表面更加光滑,骨小梁大部分排列致密整齐,小部分区域出现萎缩,稀疏,断裂.结论 EPO可以降低空骨陷窝率,增加股骨头供血微血管数量,降低血液中的脂质含量,可以预防股骨头坏死的发生和进展.

목적 관찰중조인촉홍세포생성소(EPO)대대서고골두배사적예방작용.방법 장웅성SD대서60지수궤분위3조,분별위공백조、대조조、실험조.장대조조재분위조A화조B:조A매주일、주오재대서둔기처주사작산발니송룡50 mg/kg,1차/천,지속4주;조B재진행조모지전3h재대서둔기처주사동EPO등체적적0.9%록화납주사액,기여처리조시동조A.실험조매차조모전3h재대서둔기처주사EPO용액,5000 IU/kg.결과 (1)골함와솔:소목소-이홍(HE)염색절편현시실험조공골함와솔위(14.70±1.78)%,대조조위(23.21±2.34)%,공백조위(7.45±1.23)%;(2)혈액중지질함량:분별우1、2、4주검측혈액중삼선감유함량결과여하:공백조위(1.25±0.32)、(1.28 ±0.26)、(1.27±0.40) mmo/L;대조조위(2.17±0.23)、(2.48±0.41)、(1.88 ±0.38) mmo/L;실험조위(1.42±0.34)、(1.47±0.26)、(1.50±0.49) mmo/L.총담고순함량분별재1、2、4주변화:공백조:(1.62±0.13)、(1.62±0.14)、(1.62±0.19) mmo/L;대조조:(2.41±0.18)、(2.52±0.24)、(2.13±0.37) mmo/L;실험조:(1.64±0.18)、(1.69±0.24)、(1.66±0.31)mmo/L.(3)조직학검측:실험조여대조조비교,고골두표면경가광활,골소량대부분배렬치밀정제,소부분구역출현위축,희소,단렬.결론 EPO가이강저공골함와솔,증가고골두공혈미혈관수량,강저혈액중적지질함량,가이예방고골두배사적발생화진전.
Objective To explore the preventive effect of recombinant human erythropoietin erythropoietin (EPO) on femoral head necrosis in rats.Methods Sixty male SD rats were randomly into three groups:blank group,control group,and experimental group.The control group was subdivided into two subgroups:subgroup A and subgroup B.The rats in subgroup A were injected with prednisone acetate 50 mg/kg in the root of the gluteal muscle on every Monday and Friday,once per day for 4 weeks,and those in subgroup B were injected with the same volume of sodium chloride injection of recombinant human erythropoietin erythropoietin (rhEPO,5 000 U/kg)three h before the modeling.Results The empty lacunae rate of hematoxylin and eosin (HE) stained sections in the experimental group,control group and blank group was (14.7 ± 1.78) %,(23.21 ± 2.34) %,and (7.45 ± 1.23) % respectively;At 1 st,2nd and 4th,blood triglyceride content was (1.25 ± 0.32),(1.28 ± 0.26) and (1.27 ± 0.4) mmo/L in blank group,(2.17 ±0.23),(2.48 ±0.41) and (1.88 ±0.38) mmo/L in control group,and (1.42 ±0.34),(1.47 ±0.26) and (1.50 ± 0.49) mmo/L in experimental group;the total cholesterol content was (1.62 ±0.13),(1.62 ±0.14) and (1.62 ±0.19) mmo/L in blank group,(2.41 ±0.18),(2.52±0.24) and (2.13 ±0.37) mmo/L in control group,and (1.64 ±0.18),(1.69 ±0.24) and (1.66 ± 0.31) mmo/L in experimental group.Histologically,the femoral head surface was more smooth in experimental group than control group.In experimental group,most of the trabecular bones arranged densely and regularly,and shrink,sparse,and fracture were seen in less regions.Conclusion EPO can reduce the empty lacunae rate,increase the microvessels in femoral head blood supply,reduce blood lipid levels,and prevent the progress of femoral head necrosis.
