目的探讨趋化因子CXCL5在胃癌中的表达及其与临床病理特征及预后的关系,分析其在肿瘤发生发展、侵袭转移中的作用.方法收集260例胃癌组织标本,其中60例收集血清标本.采用免疫组织化学法检测胃癌组织和正常黏膜中CXCL5蛋白表达,采用酶联免疫吸附试验法检测患者和正常志愿者血清中CXCL5含量,并分析其与临床病理特征及预后的关系.结果 CXCL5蛋白在胃癌组织中的阳性表达率[37.69％ (86/260)]高于周围正常组织[6.53％ (17/260),P＜0.01];癌组织中CXCL5表达与肿瘤大小(P＜ 0.05)、病理类型(P＜0.05)、脉管癌栓(P＜0.05)、T分期(P＜0.05)、N分期(P＜0.05)明显相关;CXCL5表达是胃癌患者的独立预后因素(P＜0.05).胃癌患者血清CXCL5的含量(985.46 ng/L)明显高于正常对照(628.49 ng/L,P＜0.01),且与肿瘤大小(P＜0.05)、脉管癌栓(P＜0.05)、T分期(P＜0.05)、M分期(P＜0.05)明显相关.结论 CXCL5在胃癌中高表达,与胃腺癌的临床分期、病理分级呈正相关,与患者生存率负相关.CXCL5可能在胃癌的发生发展、侵袭转移发挥作用.

Objective To explore the expression ofchemokine (C-X-C motif) ligand (CXCL)5 in patients with gastric cancer and analysize the roles of CXCL5 in the development,invasion and metastasis of gastric cancer.Methods The tissue specimens of 260 patients surgically treated in the Department of General Surgery,Affiliated Tumor Hospital of Zhengzhou University were collected,and 60 serum samples were collected.The expression of CXCL5 protein in tumor tissues and case-paired normal tissue samples was detected by immunohistochemistry (IHC).The concentration of serum CXCL5 in 60 cases of gastric cancer and 25 healthy volunteers was measured by enzyme linked immunosorbent assay (ELISA).Results The IHC revealed that the positive expression rate of CXCL5 was significantly higher in tumor tissues [37.69％ (86/260)] than in corresponding normal tissues [6.53％ (17/260),P ＜ 0.01].The expression of CXCL5 was correlated with tumor size (P ＜ 0.05),pathological types (P ＜ 0.05),intravascular cancer emboli (P ＜ 0.05),T staging (P ＜ 0.05) and N staging (P ＜ 0.05).The expression of CXCL5 was the independent risk factor of poor prognosis in patients with gastric cancer (P ＜ 0.05).The concentration of serum CXCL5 in gastric cancer patients (985.46 ng/L) was higher than in healthy controls (628.49 ng/L,P ＜ 0.01).High level of CXCL5 was correlated with tumor size (P ＜ 0.05),intravascular cancer emboli (P ＜ 0.05),T staging (P ＜ 0.05) and M staging (P ＜ 0.05).Conclusion The over-expression and increased concentration of CXCL5 were associated with clinical staging,pathological grade and survival time.CXCL5 may play an important role in the development,invasion and metastasis of gastric cancer.
