采集了23种鳅科鱼类89尾个体的线粒体COI基因5端序列,分析了碱基组成以及不同鳅科鱼类之间的种间遗传距离和种内遗传距离。结果显示,鳅科鱼类的种间遗传距离显著大于种内遗传距离,表明以COI作为鳅科鱼类DNA条形码进行品种鉴定具有一定的可行性。在建立鳅科鱼类DNA条形码的基础上,设计了主要物种泥鳅(Misgurnus anguillicaudatus)和大鳞副泥鳅(Paramisgurnus dabryanus)特异性引物,结果显示,该引物具有较高的特异性和灵敏度,能够实现对泥鳅、大鳞副泥鳅及其混合样品的物种鉴定。

Mitochondrial COI 5 ’ sequences of 89 cobitidae fishes belonging to 23 species were collected .The DNA base composition was analyzed , the genetic variation among species and within species were calculated .It was proved that COI gene is a valid DNA barcoding gene for species identification of Cobitidae .On the basis of Cobitidae DNA barcoding, spe-cies-specific primers for Misg runus anguillicaudatus and Para misgurnus dabryanus were designed.The result suggests the species-specific primers could identify Misgurnus anguillicaudatus, Paramisgurnus dabryanus and their mixtures with high specificity and sensitivity .
