目的:观察人抗原R( HuR)和缺氧诱导因子( HIF)-1α在肝癌组织中的表达变化,并探讨其相关性。方法取48例肝癌组织及癌旁组织标本,采用免疫组化染色法检测肝癌组织及癌旁组织中HuR和HIF-1α蛋白的表达,采用荧光定量PCR法检测肝癌组织中HIF-1αmRNA的表达。结果肝癌组织中HuR和HIF-1α蛋白阳性表达率分别为67%和58%,显著高于癌旁组织的19%和10%( P均<0.01)。肝癌组织中HuR与HIF-1α蛋白的表达呈正相关(r=0.723,P<0.01)。 HuR蛋白高表达者中HIF-1αmRNA的表达较HuR蛋白低表达者显著上调(P<0.05)。结论肝癌组织中HuR和HIF-1α蛋白表达上调且呈正相关关系,HuR可能增强HIF-1αmRNA的稳定性,从而促进其表达。

목적:관찰인항원R( HuR)화결양유도인자( HIF)-1α재간암조직중적표체변화,병탐토기상관성。방법취48례간암조직급암방조직표본,채용면역조화염색법검측간암조직급암방조직중HuR화HIF-1α단백적표체,채용형광정량PCR법검측간암조직중HIF-1αmRNA적표체。결과간암조직중HuR화HIF-1α단백양성표체솔분별위67%화58%,현저고우암방조직적19%화10%( P균<0.01)。간암조직중HuR여HIF-1α단백적표체정정상관(r=0.723,P<0.01)。 HuR단백고표체자중HIF-1αmRNA적표체교HuR단백저표체자현저상조(P<0.05)。결론간암조직중HuR화HIF-1α단백표체상조차정정상관관계,HuR가능증강HIF-1αmRNA적은정성,종이촉진기표체。
Objective To investigate the expression and correlation of human antigen R ( HuR) and hypoxia-induc-ible factor (HIF)-1αin hepatocellular carcinoma (HCC) tissues.Methods Forty-eight cases of HCC tissue and adjacent tissue samples were collected from patients undergoing surgical resection .The expression of HuR and HIF-1αprotein in the HCC and adjacent tissues was detected by immunohistochemical staining .The mRNA expression of HIF-1αin the HCC tis-sues was detected by fluorescent quantitative PCR .Results The positive expression rates of HuR and HIF-1αprotein in the HCC tissues were 67%and 58%, respectively .These rates were significantly higher than the positive expression rates of HuR and HIF-1αprotein (19%and 10 %) in the adjacent tissues ( all P<0.01) .The expression of HuR protein was positively correlated with the expression of HIF-1αprotein in the HCC tissues (r=0.723, P<0.01).The mRNA expres-sion of HIF-1αmRNA in the group with high expression of HuR protein was significantly up-regulated as compared with that of the group with low expression of HuR protein (P<0.05).Conclusions The expression of HuR and HIF-1αprotein was up-regulated and positively correlated with each other in the HCC tissues .HuR may enhance the stability of HIF-1αmRNA, and thus increase the expression of HIF-1αprotein.
