目的:建立同时测定大鼠血清中多索茶碱和甲泼尼龙质量浓度的反相高效液相色谱(RP-HPLC)法。方法以氢化可的松为内标物,色谱柱为Diamonsil C18柱(150mm ×4.6 mm,5μm),流动相为甲醇-0.5%醋酸溶液(54:46),检测波长260 nm,柱温30℃,流速1.0 mL/min,进样量20μL。结果多索茶碱、甲泼尼龙及血清内源性物质分离良好,多索茶碱质量浓度线性范围是0.5~40μg/mL ( r=0.9995),甲泼尼龙质量浓度线性范围是0.3~24μg/mL( r=0.9996),两者日内和日间 RSD均小于10%( n=5),平均回收率分别为93.49%( RSD=2.88%)和91.10%( RSD=2.56%)。结论该法简便、灵敏、准确,可用于多索茶碱和甲泼尼龙药代动力学、药物相互作用的研究和临床治疗药物监测。

Objective To establish a RP-HPLC method for the simultaneous determination of doxofylline and methylprednisolone con-tents in rat serum. Methods Hydrocortisone was selected as the internal standard. The chromatographic separation of analytes was achieved on the Diamonsil C18 column(150mm × 4. 6 mm,5 μm) at the temperature of 30 ℃, using the mobile phase consisting of methanol-0. 5% acetic acid solution ( 54 :46 ) at a flow rate of 1. 0 mL/min . The detection wavelength was set at 260 nm. Results Doxofylline, methylprednisolone and endogenous substances in serum were separated well. The linear ranges of doxofylline and methylpred-nisolone were 0. 5-40 μg/mL ( r=0. 999 5 ) and 0. 3-24 μg/mL ( r=0. 999 6 ) . The inter-day and intra-day RSD of doxofylline and methylprednisolone were all less than 10% ( n=5 ) . The average recovery rates were 93. 49% ( RSD=2. 88% ) for doxofylline and 91. 10%( RSD=2. 56%) for methylprednisolone. Conclusion This method can simultaneously determine the serum concentrations of dox-ofylline and methylprednisolone, which is convenient, sensitive, accurate and can be used to study the pharmacokinetic interaction and monitoring of serum drug concentration.
