白血病干细胞与白血病的发生、耐药及复发有关,要根除白血病,应考虑以白血病干细胞为治疗靶点.然而,白血病干细胞与正常造血干细胞具有很多相似特点,如何根除白血病干细胞而不影响正常造血干细胞的功能给研究者们提出了巨大挑战.近年研究发现,磷酸酯酶与张力蛋白同源物(PTEN)对造血干细胞与白血病干细胞的影响机制不同,这为白血病的靶向治疗带来机会.笔者拟就近年来有关PTEN对白血病干细胞及造血干细胞的影响机制及调节机制研究进行综述,旨在为白血病的靶向治疗提供新思路.
白血病榦細胞與白血病的髮生、耐藥及複髮有關,要根除白血病,應攷慮以白血病榦細胞為治療靶點.然而,白血病榦細胞與正常造血榦細胞具有很多相似特點,如何根除白血病榦細胞而不影響正常造血榦細胞的功能給研究者們提齣瞭巨大挑戰.近年研究髮現,燐痠酯酶與張力蛋白同源物(PTEN)對造血榦細胞與白血病榦細胞的影響機製不同,這為白血病的靶嚮治療帶來機會.筆者擬就近年來有關PTEN對白血病榦細胞及造血榦細胞的影響機製及調節機製研究進行綜述,旨在為白血病的靶嚮治療提供新思路.

Leukemia stem cells are considered responsible for the occurrence,prognosis,relapse and resistance to chemotherapy of leukemia.In order to completely eradicate leukemia cells and consequently cure the disease,leukemia stem cells therapies should be considered as a therapeutic target.However,the fact that leukemia stem cells share functional and phenotypic properties with normal hematopoietic stem cells poses a significant challenge to researchers:how to definitely eradicate leukemia stem cells without damaging normal hematopoietic stem cells and compromising hematopoiesis.In recent years,researchers found that phosphatase and tensin homolog (PTEN) regulates leukemia stem cells and hematopoietic stem cells through different mechanisms.And this discovery demonstrates that it is possible to identify pathways that differentially impact leukemia and normal stem cell function and opens new therapeutic perspectives for the selective elimination of leukemia stem cells.In this review,we briefly discuss the mechanisms that regulate PTEN function in leukemia stem cells and hematopoietic stem cells and their potential for the development of leukemia stem cells-targeted therapies.
