为了优化甘蓝型油菜小孢子培养技术,提高胚诱导率及成苗率,以油菜N14009为试验材料进行小孢子培养,研究变温预处理、活性炭浓度、培养基类型和琼脂浓度及秋水仙碱浓度对小孢子培养的影响。结果表明:4℃、32.5℃变温预处理各1 d的处理效果较好,胚诱导率为4.23胚/蕾;活性炭质量浓度为0.5 g/L时,胚诱导率达到4.23胚/蕾,效果最好; MS、B5培养基中琼脂质量浓度为12 g/L时,成苗率均最高,分别达51.16%、46.67%;把再生植株转到含有不同质量浓度秋水碱的MS培养基上培养,质量浓度为80 mg/L时,N14009的加倍率最高,为43.47%,并且成苗率达到76.67%。

In order to optimize microspore culture techniques of Brassica napus L. and improve the embryo induction rate and seedling rate, the N14009 was used as experimental material to culture microspores. During the research,the effects on microspore culture were studied by different temperature pretreatment,concentration of activated carbon,medium type and agar concentration and concentration of colchicine . The results showed that:It got a good result by 4℃ and 32 . 5℃ temperature pretreatments for one day each,and the embryo induction rate was 4. 23 embryos/bud;When the concentration of activated carbon was 0. 5 g/L,the embryo induction rate reached the maximum,4. 23 embryos/bud;In addition,the seedling rate reached 51. 16% and 46. 67% in the MS and B5 media both with 12 g/L of agar, respectively. The regenerated plants were turned to MS that had different concentration of colchicine, the doubling efficiency of N14009 reached 43 . 47% and the seedling rate reached 76 . 67%, when the concentration of colchicine was 80 mg/L.
