目的:观察骨髓间充质干细胞的条件培养基(BMSC-CM)局部移植对脊髓损伤修复的作用。方法采用改良的Allen 装置制作大鼠 T9脊髓损伤模型,随机分为对照组(脊髓损伤后局部微量注射生理盐水10μl)和实验组(脊髓损伤后局部注射 BMSC-CM 10μl),两组均采用 BBB 评分、斜板实验法观察大鼠运动功能恢复情况,苏木精-伊红(HE)染色观察脊髓损伤处血管生长及空洞体积的改变情况。结果实验组大鼠的 BBB 后肢功能评分、斜板倾斜角度优于对照组(P <0.05),脊髓损伤处空洞体积明显小于对照组,血管直径大于对照组。结论 BMSC-CM 能够减小脊髓损伤处的空洞体积和促进血管生长,从而促进脊髓损伤后神经功能的恢复。
目的:觀察骨髓間充質榦細胞的條件培養基(BMSC-CM)跼部移植對脊髓損傷脩複的作用。方法採用改良的Allen 裝置製作大鼠 T9脊髓損傷模型,隨機分為對照組(脊髓損傷後跼部微量註射生理鹽水10μl)和實驗組(脊髓損傷後跼部註射 BMSC-CM 10μl),兩組均採用 BBB 評分、斜闆實驗法觀察大鼠運動功能恢複情況,囌木精-伊紅(HE)染色觀察脊髓損傷處血管生長及空洞體積的改變情況。結果實驗組大鼠的 BBB 後肢功能評分、斜闆傾斜角度優于對照組(P <0.05),脊髓損傷處空洞體積明顯小于對照組,血管直徑大于對照組。結論 BMSC-CM 能夠減小脊髓損傷處的空洞體積和促進血管生長,從而促進脊髓損傷後神經功能的恢複。
목적:관찰골수간충질간세포적조건배양기(BMSC-CM)국부이식대척수손상수복적작용。방법채용개량적Allen 장치제작대서 T9척수손상모형,수궤분위대조조(척수손상후국부미량주사생리염수10μl)화실험조(척수손상후국부주사 BMSC-CM 10μl),량조균채용 BBB 평분、사판실험법관찰대서운동공능회복정황,소목정-이홍(HE)염색관찰척수손상처혈관생장급공동체적적개변정황。결과실험조대서적 BBB 후지공능평분、사판경사각도우우대조조(P <0.05),척수손상처공동체적명현소우대조조,혈관직경대우대조조。결론 BMSC-CM 능구감소척수손상처적공동체적화촉진혈관생장,종이촉진척수손상후신경공능적회복。
Objective To investigate the promoting effects of bone marrow mesenchymal stem cells conditioned me-dium (BMSC-CM) local transplantation on function recovery of rat spinal cord injuries. Methods The spinal cord of adult rats was injured with modified Allen method, and the rats were randomly divided into 2 groups. The rats in the control group were subjected to the microinjection of saline solution (10 μl) into the spinal cord injuries site af-ter spinal cord injuries, and those in the experimental group were injected with the concentrated conditioned medi-um (10 μl) derived from the passage 3 BMSCs. The inclined plane test and Basso, Beattie and Bresnahan locomo-tor rating scale (BBB scale) were used to observe the recovery of rats’behavioral function after the transplantation. Hematoxylin-eosin (HE) staining to reflect the change of cavity volume and angiogenesis at the lesion epicenter. Results BMSC-CM treated animals showed better BBB scores and inclined plane scores than control animals, and this difference was statistically significant (P < 0. 05). BMSC-CM treated group showed reduced cavity volume and the blood vessels exhibited larger diameters compared to the control group. Conclusion BMSC-CM can promote the recovery of neurological function in spinal cord injury, reduce cavity volume, and improve blood vessel growth.
