目的:探讨关于鼠肾小管上皮细胞原代培养的有效方法。方法在现有肾小管上皮细胞机械研磨方法的基础上,通过SD大鼠与昆明小鼠的对比实验,Ⅰ型胶原酶消化时间的设定以及第一次换取液的合理利用等方面改进原代培养的方法。对改进后所分离的肾小管上皮细胞进行鉴定。结果与SD大鼠相比,昆明小鼠细胞贴壁生长多且快,胶原蛋消化25分钟后的细胞贴壁多、长满壁底早。将第一次细胞换取液进行再次贴壁,第二天依旧可以有部分细胞贴壁,3~4天长满壁底,纯度较高。两次细胞均成上皮细胞鹅卵石铺路样生长,经免疫细胞化学法(SABC法)鉴定CK表达阳性。结论实验选择昆明小鼠肾小管上皮细胞经胶原酶消化25分钟左右的细胞数量、质量较好,第一次细胞废弃液可以再次有效利用。

Objective:To explore the effective methods for the primary culture of rat renal tubular epithelial cells. Methods:On the basis of the mechanical grinding for the obtainment of tubular epithelial cells, we compared the difference between the experimental SD rats and KunMing mice as the source and optimized theⅠcollagenase digestion time as well as the rational use of liquid that was quitted for the ifrst time. The identiifcation of the collected renal tubular epithelial cells was performed by immunocytochemistry. Results:Compared with SD rats, renal tubular epithelial cells isolated from KunMing mice presented better growth rate with 25 minutes’ Ⅰcollagenase digestion. Additionally, the amount and quality of cells which recovered from the ifrst cell waste liquid were good, showing an effective reutilization. Conclusion:Mechanical grinding and 25 minutes’Ⅰcollagenase digestion with KunMing mice as source is an ideal method for the primary culture of the renal tubular epithelial cells. And the ifrst cell waste liquid could be reused well.
