目的:观察中老年部分雄激素缺乏综合征(PADAM)大鼠睾丸细胞色素P450侧链裂解酶(P450scc)和类固醇生长因子-1(SF-1)的表达及电针干预对其表达的影响,探讨P450scc和SF-1在电针调节睾酮分泌过程中的作用。方法将40只雄性SD大鼠分为正常组(n=10)和造模组(n=30),造模组腹腔注射环磷酰胺复制PADAM模型。从造模成功的大鼠中随机选取20只分为电针组(n=10)和模型组(n=10)。电针组取“肾俞”“关元”针灸,留针15 min,每日治疗1次。在第8周用ELISA分别检测正常组、电针组和模型组大鼠血清的总睾酮(TT)、游离睾酮(FT)水平;免疫组织化学法检测P450scc蛋白表达;Western印迹检测SF-1蛋白表达;RT-PCR检测P450scc和SF-1mRNA的表达。结果治疗8 w时,ELISA结果显示,电针组血清TT、FT水平明显高于模型组( P<0.01);电镜结果显示,电针组Leydig细胞正常,线粒体和内质网含量较模型组多;HE染色结果显示,电针组睾丸组织中曲细精管管腔肿胀程度较模型组减轻,Sertoli细胞较模型组排列整齐;免疫组化和WB结果显示电针组P450scc和SF-1蛋白表达较模型组升高(P<0.01);RT-PCR结果显示,电针组P450scc和SF-1mRNA表达较模型组升高(P<0.01),且两者具有相关性(|r|≥0.8,P<0.01)。结论电针能明显升高PADAM大鼠体内睾酮水平,改善睾丸Leydig细胞和Sertoli细胞病理表现。电针干预后可提高P450scc和 SF-1的表达,这可能是电针治疗PADAM的作用机制之一。

Objective To observe the effect of electroacupuncture ( EA) intervention on expression of cytochrome P450 side chain cleavage (P450scc) and steroidogenic factor(SF)-1 in the testis of partial androgen deficiency of aging male (PADAM) rats so as to study its mechanism to improve testosterone.Methods A total of 40 male Sprague Dawley rats were randomly divided into control ( n=10 ) and building model groups(n=30),and building model group'rats were injected in abdominal cavities with cyclophosphamide to make PADAM model.PADAM model rats were randomized into EA and model groups.The serum TT and FT levels were measured by enzyme-linked immu-nosorbent assay ( ELISA) .The expression levels of P450scc and SF-1 proteins and mRNA in testis were assayed by immunohistochemistry, Western blot and RT-PCR.Results After 8 weeks,the serum TT,FT levels in EA group were significantly higher than those in model group (P<0.01).The expression levels of P450scc and SF-1 proteins and mRNA in EA group were significantly higher than those in model group (P<0.01).Conclusions EA is an effective method to improve serum TT,FT levels of PADAM rats.It also could significantly improve tes-ticular P450scc and SF-1 proteins and mRNA,which might be one of the mechanisms of EA therapy for PADAM.
