目的：探讨重组人白细胞介素（ IL）-6对胶质瘤细胞U87增生、侵袭能力的影响及其机制。方法应用CCK-8法检测IL-6对U87细胞增生的影响并计算增殖率；应用Transwell小室检测IL-6对U87侵袭的影响；应用Western印迹检测IL-6对胶质瘤细胞STAT3蛋白及其磷酸化状态的影响。结果 IL-6能促进U87增生和侵袭能力（P＜0.05），并伴随STAT3及p-STAT3蛋白表达上调（P＜0.05）。结论 IL-6通过促进胶质瘤细胞STAT3磷酸化，从而促进了胶质瘤细胞的增生侵袭能力，为脑胶质瘤的临床治疗提供了理论支持。
목적：탐토중조인백세포개소（ IL）-6대효질류세포U87증생、침습능력적영향급기궤제。방법응용CCK-8법검측IL-6대U87세포증생적영향병계산증식솔；응용Transwell소실검측IL-6대U87침습적영향；응용Western인적검측IL-6대효질류세포STAT3단백급기린산화상태적영향。결과 IL-6능촉진U87증생화침습능력（P＜0.05），병반수STAT3급p-STAT3단백표체상조（P＜0.05）。결론 IL-6통과촉진효질류세포STAT3린산화，종이촉진료효질류세포적증생침습능력，위뇌효질류적림상치료제공료이론지지。
Objective To study the effect of recombinant human interleukin( IL)-6 on proliferation and invasion of Human U87 gli-oma cells and its mechanism.Methods CCK-8 assay was used to detect the effect of IL-6 on proliferation of U87 glioma cells,and the rela-tive proliferation effect was calculated.Transwell assay was conducted to study the effect of IL-6 on invasion of U87 glioma cells.Western blot was performed to investigate the change of Stat3 protein and its phosphorylation status after IL-6 treatment.Results The IL-6 promoted the proliferation of U87 glioma cells(P<0.05).The invasion abilities of U87 cells were significantly promoted by IL-6 treatment(P<0.05), which was accompanied by the up-regulation of the STAT3 and p-STAT3 protein expressions.Conclusions IL-6 could effectively promote U87 glioma cells proliferation and invasion.The phosphorylation of STAT3 might play an important role in this process.
