目的 探讨青岛地区汉族男性群体的IL-13基因第四外显子区域+2044C/T(rs20541)多态性与原发性痛风遗传易感性的关系.方法 采用TaqMan荧光定量PCR技术,对青岛地区281例男性原发性痛风患者与265名男性对照的+2044C/T位点进行基因分型.采用Hardy-Weinberg检验确认标本的群体代表性,数据分析采用x2检验和t检验.结果 痛风组中IL-13 +2044C/T位点CC,CT和TT基因型分别为82例、146例和53例(29.2%,52.0%和18.9%),对照组分别为92名、127名和48名(34.7%,47.9%和18.1%),2组比较差异无统计学意义(x2=1.788,P>0.05);2组的等位基因频率C和T间差异也无统计学意义(分别为55.2%,44.8%;58.2%,41.8%;x2=1.057,P>0.05).结论 尚不能认为青岛地区汉族男性人群中IL-13 +2044C/T位点基因多态性与痛风有关联性.

Objective To explore gene polymorphism of the C/T genotype of +2044C/T in the fourth exon of IL-13 gene in males of Qingdao,and thus to investigate the relationship between the gene polymorphism of IL-13 and primary gout.Methods A total of 281 gout patients and 265 controls were enrolled.The possible association between the polymorphism of IL-13 +2044C/T and gout in Qingdao was investigated and genotype frequencies and allelic frequencies were calculated by real-time polymerase chain reaction (RT-PCR) with Taqman probe method.Hardy-Weinberg was used to verify the representativeness of the sample.Comparison between the groups was performed with x2 test and t-test.Results The frequencies of CC,CT and TT genotypes were 82,146 and 53 (29.2%,52.0% and 18.9%) respectively among gout patients,while they were 92,127 and 48 (34.7%,47.9% and 18.1%),respectively among the controls.There was no statistical significant difference in IL-13 +2044C/T genotype frequencies between gout patients and controls (x2=1.788,P>O.05).The allele frequencies of C and T in gout patients were different from those in the controls (55.2%,44.8%;58.2%,41.8%;x2=1.057,P>0.05).Conclusion Results of the present study suggest that the C/T genotype of +2044C/T in the fourth exon of IL-13 gene is not associated with gout in males of Qingdao.
