目的:探讨参苓白术散对脾虚湿困型溃疡性结肠炎( UC)大鼠结肠组织NF-κB p65蛋白表达及相关炎性因子的影响。方法将48只Wistar大鼠按照随机数字表分为正常组、模型组、柳氮磺吡啶组、参苓白术散组,每组12只,脾虚湿困型溃疡性结肠炎大鼠模型采用三硝基苯磺酸( trinitrobenzene sulfonic acid,TNBS)/乙醇灌肠结合环境与饮食干预法复制,运用免疫组织化学法检测并比较各组大鼠结肠组织NF-κB p65蛋白表达,ELISA法测定血清IL-6、IL-8、TNF-α水平的变化。结果模型组大鼠NF-κB p65蛋白表达及IL-6、IL-8、TNF-α水平明显高于正常组( P<0.05),参苓白术散组和柳氮磺吡啶组较模型组显著降低,差异有统计学意义(P<0.05),参苓白术散组与柳氮磺吡啶组比较差异无统计学意义(P>0.05)。结论参苓白术散可以明显改善UC大鼠结肠组织中NF-κB p65蛋白表达及血清IL-6、IL-8、TNF-α水平,促进肠黏膜的修复。
目的:探討參苓白術散對脾虛濕睏型潰瘍性結腸炎( UC)大鼠結腸組織NF-κB p65蛋白錶達及相關炎性因子的影響。方法將48隻Wistar大鼠按照隨機數字錶分為正常組、模型組、柳氮磺吡啶組、參苓白術散組,每組12隻,脾虛濕睏型潰瘍性結腸炎大鼠模型採用三硝基苯磺痠( trinitrobenzene sulfonic acid,TNBS)/乙醇灌腸結閤環境與飲食榦預法複製,運用免疫組織化學法檢測併比較各組大鼠結腸組織NF-κB p65蛋白錶達,ELISA法測定血清IL-6、IL-8、TNF-α水平的變化。結果模型組大鼠NF-κB p65蛋白錶達及IL-6、IL-8、TNF-α水平明顯高于正常組( P<0.05),參苓白術散組和柳氮磺吡啶組較模型組顯著降低,差異有統計學意義(P<0.05),參苓白術散組與柳氮磺吡啶組比較差異無統計學意義(P>0.05)。結論參苓白術散可以明顯改善UC大鼠結腸組織中NF-κB p65蛋白錶達及血清IL-6、IL-8、TNF-α水平,促進腸黏膜的脩複。
목적:탐토삼령백술산대비허습곤형궤양성결장염( UC)대서결장조직NF-κB p65단백표체급상관염성인자적영향。방법장48지Wistar대서안조수궤수자표분위정상조、모형조、류담광필정조、삼령백술산조,매조12지,비허습곤형궤양성결장염대서모형채용삼초기분광산( trinitrobenzene sulfonic acid,TNBS)/을순관장결합배경여음식간예법복제,운용면역조직화학법검측병비교각조대서결장조직NF-κB p65단백표체,ELISA법측정혈청IL-6、IL-8、TNF-α수평적변화。결과모형조대서NF-κB p65단백표체급IL-6、IL-8、TNF-α수평명현고우정상조( P<0.05),삼령백술산조화류담광필정조교모형조현저강저,차이유통계학의의(P<0.05),삼령백술산조여류담광필정조비교차이무통계학의의(P>0.05)。결론삼령백술산가이명현개선UC대서결장조직중NF-κB p65단백표체급혈청IL-6、IL-8、TNF-α수평,촉진장점막적수복。
Objective To investigate the effects of Shenlin Baizhu San ( Ginseng, Poria and Atractylodes Macrocephalae Powder) on the protein expression of NF-κB p65 in the colon tissue of rats with ulcerative colitis due to dampness retention and spleen deficiency, as well as the related inflammatory cytokines. Methods 48 Wistar rats were divided into four groups: normal group, model group, sulfasalazine group, and Shenlin Baizhu San group, with 12 rats in each group.The model rats were induced by environment and diet intervention combined with composite trinitrobenzene sulfonic acid and ethanol in enema.The protein expression of NF-κB p65 in the colon tissue of rats were detected with immunohisto-chemistry methods.Levels of IL-6,IL-8, and TNF-αwere measured with ELISA.Results The protein expression of NF-κB p65 and the serum level of IL-6, IL-8, and TNF-αin the model rats were significantly higher than those in normal group ( P<0 .05 ) .The protein expression of NF-κB p65 and the serum level of IL-6, IL-8, and TNF-αin Shenlin Baizhu San and sulfasalazine groups significantly decreased, and the difference was significant compared with model group (P<0.05).The difference between Shenlin Baizhu San and sulfasalazine groups showed no statistical significance ( P >0.05 ) . Conclusion Shenlin Baizhu San could significantly improve the protein expression of NF-κB p65 and the serum level of IL-6,IL-8, and TNF-αin the colon tissue of rats with ulcerative colitis, which could promote the repair of the intestinal mucosa.
