为研究125 I‐rAncrod（1.59μg · kg －1）在Wistar大鼠器官和组织分布以及粪尿和胆汁的代谢情况，采用放射性125 I标记重组安克洛酶（125 I‐rAncrod）结合分子排阻色谱法（size exclusion chromatography ， SEC）及γ‐计数法测定不同时间组织及体液样品中125 I‐rAncrod的含量。结果显示，放射性药物浓度主要累积在甲状腺和胃肠道内，肝肾次之，而脑、脊髓、脂肪、肌肉中药物浓度一直处于较低的水平。Wist‐ar大鼠尾静脉注射125 I‐rAncrod后，SEC色谱图显示到48 h粪尿和胆汁中均检测不到原形药物。结果表明，大鼠尾静脉注射125 I‐rAncrod后在全身各器官和组织分布广泛，代谢较完全。
위연구125 I‐rAncrod（1.59μg · kg －1）재Wistar대서기관화조직분포이급분뇨화담즙적대사정황，채용방사성125 I표기중조안극락매（125 I‐rAncrod）결합분자배조색보법（size exclusion chromatography ， SEC）급γ‐계수법측정불동시간조직급체액양품중125 I‐rAncrod적함량。결과현시，방사성약물농도주요루적재갑상선화위장도내，간신차지，이뇌、척수、지방、기육중약물농도일직처우교저적수평。Wist‐ar대서미정맥주사125 I‐rAncrod후，SEC색보도현시도48 h분뇨화담즙중균검측불도원형약물。결과표명，대서미정맥주사125 I‐rAncrod후재전신각기관화조직분포엄범，대사교완전。
To study the tissue distribution and metabolism of radioactivity in rats after single intravenous administration of 125 I‐rAncrod(1.59μg · kg -1 ) .The concentration of 125 I‐rAncrod in various tissue and body fluids were determined by 125 I labeled rAncrod combined with size exclusion chromatography (SEC) andγ‐counting methods .The con‐centration of 125 I‐rAncrod predominantly accumulated in the thyroid and gastrointestinal and also be found in the kidney and liver in fair amounts ,but in brain ,spinal cord , muscle and fat was at a very low level .After single intravenous administration (chemi‐cal dose 1.59 μg · kg -1 ) of 125 I‐rAncrod to Wistar rats ,the samples of feces ,urine and bile were analyzed by SEC .The result was that parent drug was not existed in feces , urine and bile in 48 h .The results showed that 125 I‐rAncrod was distributed widely in rats ,metabolic process was complete .
