采用变性梯度凝胶电泳技术研究青海农村户用沼气池微生物群落,对其PCR-DGGE的电泳时间及上样量进行优化。结果表明:最佳电泳时间为16 h,最适上样量为4μg,优化后的PCR-DGGE确保了实验的准确性和可重复性。运用此优化后的PCR-DGGE技术对2个户用沼气池内细菌和古菌群落进行了研究,获得了较丰富的多样性。

The microbial community in rural household biogas digesters of Qinghai was investigated in this study with PCR -DGGE technology .In order to improve the accuracy and repeatability of DGGE, the running parameters of DGGE were optimized by electrophoresis time and sampling a-mount, respectively.The results showed that the optimal running time was 16 hours, and the sam-pling amount was 4μg.Furthermore, bacterial community and archaeal community from two rural household biogas digesters were also performed by using these optimized reaction conditions .It was found that the diversity of bacteria and archaea were rich .
