目的:探讨直接覆盖玻璃化冷冻(direct cover vitrification, DCV)在冷冻大鼠卵巢组织中的效果,并对自体异位移植后卵巢组织存活状况及内分泌情况进行分析。方法性成熟的 Wistar 大鼠50只随机分为 A、B、C、D 组, A 和 B 组各20只,C 和 D 组各5只。 A 组为新鲜卵巢组织移植;B 组卵巢组织经 DCV 冷冻保存2周复苏后移植;C 组为去势对照;D 组为假手术对照。自体异位移植6周后检测大鼠血清中雌二醇(E2)水平,计算卵泡密度,观察移植卵巢组织形态学及增殖细胞核抗原(proliferating cell nuclear antigen, PCNA)的表达情况。结果大鼠卵巢组织移植存活的卵巢组织周围可见血管供应,移植失败的卵巢组织呈纤维化状态,与周围组织血供不明显。 B 组卵巢组织移植存活率低于 A 组(P <0.05)。 B 组卵巢组织中卵泡密度均低于 A、D 组,且 A 组低于 D 组(P <0.05)。 A、B 组血清 E2水平高于 C 组,低于 D 组(P <0.05)。 A、B 组卵巢组织 PCNA 阳性率均低于 D 组,且 B 组低于 A 组(P <0.05)。结论大鼠卵巢组织经 DCV 法冻融自体异位移植后部分卵泡能存活并可恢复内分泌功能。

목적:탐토직접복개파리화냉동(direct cover vitrification, DCV)재냉동대서란소조직중적효과,병대자체이위이식후란소조직존활상황급내분비정황진행분석。방법성성숙적 Wistar 대서50지수궤분위 A、B、C、D 조, A 화 B 조각20지,C 화 D 조각5지。 A 조위신선란소조직이식;B 조란소조직경 DCV 냉동보존2주복소후이식;C 조위거세대조;D 조위가수술대조。자체이위이식6주후검측대서혈청중자이순(E2)수평,계산란포밀도,관찰이식란소조직형태학급증식세포핵항원(proliferating cell nuclear antigen, PCNA)적표체정황。결과대서란소조직이식존활적란소조직주위가견혈관공응,이식실패적란소조직정섬유화상태,여주위조직혈공불명현。 B 조란소조직이식존활솔저우 A 조(P <0.05)。 B 조란소조직중란포밀도균저우 A、D 조,차 A 조저우 D 조(P <0.05)。 A、B 조혈청 E2수평고우 C 조,저우 D 조(P <0.05)。 A、B 조란소조직 PCNA 양성솔균저우 D 조,차 B 조저우 A 조(P <0.05)。결론대서란소조직경 DCV 법동융자체이위이식후부분란포능존활병가회복내분비공능。
Objective To investigate the effect of direct cover vitrification (DCV) method in freezing ovarian tissues and to analyze the viability and endocrinic function after the autologous heterotopic transplantation in rats. Meth-ods A total of 50 mature female rats were randomly divided into group A (n = 20), group B (n = 20), group C (n =5) and group D ( n = 5). Group A was transplanted with fresh tissues; group B was transplanted with cryopreserred thawed tissues after being cryopreserved for 2 weeks by DCV method; the bilateral ovarian tissues in group C were cut off, and group D underwent sham operations. After autologous heterotopic transplantation for 6 weeks, serum estradiol (E2 ) levels were detected, and values of follicular density were calculated; histomorphology changes of transplanted ovaries and expressions of proliferating cell nuclear antigen (PCNA) were observed. Results The vascularity was found in surround-ing tissues of survival ovaries, while fibering and unconspicuous vascularity were found in unsuccessfully transplanted ova-ry tissues. The survival rate of grafts in group B was lower than that in group A (P < 0. 05). The value of follicular densi-ty in group B was lower than those in group A and D, and the value in group A was lower than that in group D (P <0. 05). The serum E2 levels in group A and B were higher than that in group C, but the values were lower than that in group D (P < 0. 05). The positive rates of PCNA in group A and B were lower than that in group D, and the rate in group B was lower than that in group A (P < 0. 05). Conclusion Some ovarian tissues with autologous heterotopic transplanta-tion after DCV and resuscitation method can survive and rehabilitate cover endocrine function.
