采用 HPLC 法测定红曲霉液态发酵培养合成开环的酸型莫纳可林 K (Monacolin K)及闭环的内酯型洛伐他汀(Lovastatin)的含量,研究生长因子、前体物质和金属离子等微量营养源对红曲霉液态发酵产洛伐他汀开闭环组分的影响。结果表明:红曲霉液态发酵中,添加烟酸、乙酸钠和硫酸锌等微量营养源能促进 Monacolin K 及 Lovastatin 的合成,并提高 Monacoling K 的比例;当烟酸、乙酸钠和硫酸锌的添加量分别为:0.01%、0.15%和0.15%时,产生的开环 Monacolin K 的比例占79%,开闭环洛伐他汀总质量浓度为57.138 mg·L-1。
採用 HPLC 法測定紅麯黴液態髮酵培養閤成開環的痠型莫納可林 K (Monacolin K)及閉環的內酯型洛伐他汀(Lovastatin)的含量,研究生長因子、前體物質和金屬離子等微量營養源對紅麯黴液態髮酵產洛伐他汀開閉環組分的影響。結果錶明:紅麯黴液態髮酵中,添加煙痠、乙痠鈉和硫痠鋅等微量營養源能促進 Monacolin K 及 Lovastatin 的閤成,併提高 Monacoling K 的比例;噹煙痠、乙痠鈉和硫痠鋅的添加量分彆為:0.01%、0.15%和0.15%時,產生的開環 Monacolin K 的比例佔79%,開閉環洛伐他汀總質量濃度為57.138 mg·L-1。
채용 HPLC 법측정홍곡매액태발효배양합성개배적산형막납가림 K (Monacolin K)급폐배적내지형락벌타정(Lovastatin)적함량,연구생장인자、전체물질화금속리자등미량영양원대홍곡매액태발효산락벌타정개폐배조분적영향。결과표명:홍곡매액태발효중,첨가연산、을산납화류산자등미량영양원능촉진 Monacolin K 급 Lovastatin 적합성,병제고 Monacoling K 적비례;당연산、을산납화류산자적첨가량분별위:0.01%、0.15%화0.15%시,산생적개배 Monacolin K 적비례점79%,개폐배락벌타정총질량농도위57.138 mg·L-1。
Effect of the trace elements,including the growth factors,precursors and cations,on the production of Monacolin K and Lovastatin in the liquid fermentation of Monascus was studied.The secondary metabolites of the acidic form of Monacolin K and the lactone-form of Lovastatin were measured by using HPLC.The results showed that the addition of vitamin B3 as a growth factor,sodium acetate as a precursor and Zn2+ as a cation significantly increased the total yield of Lovastatin and raised the proportion of Monacolin K produced in the liquid fermentation of Monascus .Based on the orthogonal test,the optimized additions of these trace elements were found to be 0.15%ZnSO4 ,0.01 % vitamin B3 and 0.15% sodium acetate.The resulting total Lovastatin production was 57.138 mg·L-1 ,the acidic form of Monacolin K was 44.958 mg·L-1 ,and the proportion of the acidic form of Monacolin K increased to 79% of the total Lovastatin collected.
