目的 比较多发与单发性胶质瘤甲醛固定石蜡包埋(FFPE)组织中微小RNA (miRNA)表达谱的差异,初步探讨差异表达的miRNA及其靶基因在胶质瘤侵袭迁移中可能发挥的作用.方法 选取多发和单发胶质瘤患者FFPE标本各6例,提取总RNA并进行质检;采用基因芯片技术检测miRNA表达谱,利用芯片分析软件对结果进行数据分析,筛选出多发与单发胶质瘤表达差异有统计学意义的miRNA,再利用生物学信息库预测表达差异最显著miRNA的靶基因.结果 所有FFPE组织标本中提取的总RNA质量均符合质控标准.与单发胶质瘤相比,多发胶质瘤中共有128个miRNA表达差异有统计学意义(倍数>2或<0.5),其中有10个miRNAs显著上调(倍数>50),分别为miR-21、miR-455、miR-199、miR-410、miR-148a、miR-335、miR-34b、miR-664a、miR-542、miR-4453;有5个表达明显下调(倍数<0.05),分别为miR-200a、miR-451、miR-29a、miR-101、miR-96.基于上调和下调最显著的miR-21(差异倍数为240.11)和miR-200a(差异倍数为0.009913)靶基因预测结果,其多种靶基因涉及肿瘤侵袭迁移相关的生物学功能.结论 FFPE组织可以满足后续miRNA芯片等实验研究.多发胶质瘤与单发胶质瘤存在miRNA的差异表达,其中miR-21和miR-200a可能与胶质瘤的侵袭和迁移密切相关.

Objectives To compare the differences of microRNA (miRNA) expression profile of multiple and solitary gliomas in formalin-fixed and paraffin embedded (FFPE) tissue and to preliminarily investigate the possible roles in glioma invasion and migration for the significantly different miRNA and its target genes.Methods FFPE specimens in patients with multiple glioma (n =6) and solitary glioma (n =6) were selected.The total RNA was extracted and detected;the gene chip technology was used to detect miRNA expression profile.The chip analysis software was used to analyze the data of the results.The miRNAs of significant expression differences of the multiple and solitary gliomas were screened.Then the biological information database was used to predict the target genes of miRNA of the most significant expression differences.Results The total RNA quality extracted from all the FFPE tissue specimens met the standard of quality control.There were significant differences in 128 different miRNAs in the multiple glioma compared with the solitary glioma (multiple > 5 or < 0.5),and ten of them were significantly upregulated (multiple > 50),they were miR-21,miR-455,miR-199,miR-410,miR-148a,miR-335,miR-34b,miR-664a,miR-542,and miR-4453,respectively.Five miRNAs were significantly downregulated (multiple <0.05),they were miR-200a,miR-451,miR-29a,miR-101,and miR-96.Based on the most significant prediction results of the miR-21 and miR-200a target genes of upregulation and downregulation,their multiple target genes involved in tumor invasion and migration related biological function.Conclusions FFPE tissue may meet the subsequent experimental study,such as miRNA chip,etc.and thus provide rich resources for the research of the miRNA biological function and exploration.Multiple glioma has differential expression of miRNA compared with solitary glioma,in which miR-21and miR-200a may be closely associated with the invasion and migration of glioma.
