为快速、简便地定性定量荷叶中的黄酮类物质,建立高效液相色谱法测定荷叶中金丝桃苷、槲皮素、山奈酚和异鼠李素4种黄酮含量的方法。结果表明:在其分析测定条件色谱柱为 Gemini C18,柱温为40℃,流动相为甲醇-0.1%磷酸(55∶45,体积比),流速为1 mL/min,检测波长360 nm 的条件下,金丝桃苷、槲皮素、山奈酚和异鼠李素含量分别在1.5~48.0μg/mL,1.0~32.0μg/mL,1.0~32.0μg/mL,0.7~24.0μg/mL线性关系良好,方法的回收率为90%~103%,精密度为3.5%~4.2%,荷叶中4种黄酮含量依次为1.94%、0.092%、0.025%和0.016%。该方法可用于荷叶样品4种黄酮类物质的含量测定。

To establish a quick and easy method of qualitative and quantitative analysis for determining the content of hyperoside,quercetin,kaempferol and isorhamnetin in lotus leaves by HPLC.Results:Under conditions of Gemini C18 for the chromatographic column and 40℃ for column temperature,with methanol-0.1% phosphoric aicd(55∶45,v∶v)of the mobile phase and 1 mL/min of flow velocity,360nm for the detection wavelength,the linear ranges of hyperoside,quercetin,kaempferol and isorhamnetin were 1.5~ 48.0 μg/mL,1.0 ~ 32.0 μg/mL,1.0 ~ 32.0 μg/mL,0.7 ~ 24.0 μg/mL,respectively,the recovery was 90%~103% and the precision was 3.5%~4.2%,the contents of four kinds of flavonoids in lotus leaves were respectively 1.94%,0.092%,0.025% and 0.021%.The proposed method was suitable for the determination of hyperoside,quercetin,kaempferol and isorhamnetin in lotus leaves.
