目的:探讨亲环素 A(CyPA)/ CD147在动脉粥样硬化(AS)兔血管重构中的表达特点。方法新西兰雄性白兔32只(3月龄),随机分为对照组、模型组、液氮冻伤组、高脂饮食组(每组8只)。0周和13周分别检测兔血清 CD147和CyPA 表达水平。术后13周处死动物,取血管石蜡包埋切片,行 HE、弹力纤维染色,观察血管形态,并做巨噬细胞、CyPA、CD147免疫组织化学染色。利用计算机图像分析系统观察动脉损伤后血管重构相关指标的变化。结果13周时模型组、液氮冻伤组、高脂饮食组血 CyPA 与 CD147水平较同时期对照组均明显升高(P <0.05),尤以模型组为甚。除对照组,其余3组均可见内膜增生,模型组还可见典型 AS斑块形成;斑块内及增生内膜均可见 CyPA 及 CD147的表达。模型组、液氮冻伤组、高脂饮食组最小管腔直径及管腔面积较对照组均明显减小(P <0.05),内膜面积、中膜面积、最大内膜厚度、管腔狭窄度较对照组明显增大(P <0.05)。结论 CyPA 与 CD147在 AS 兔血管重构中的表达明显升高。

목적:탐토친배소 A(CyPA)/ CD147재동맥죽양경화(AS)토혈관중구중적표체특점。방법신서란웅성백토32지(3월령),수궤분위대조조、모형조、액담동상조、고지음식조(매조8지)。0주화13주분별검측토혈청 CD147화CyPA 표체수평。술후13주처사동물,취혈관석사포매절편,행 HE、탄력섬유염색,관찰혈관형태,병주거서세포、CyPA、CD147면역조직화학염색。이용계산궤도상분석계통관찰동맥손상후혈관중구상관지표적변화。결과13주시모형조、액담동상조、고지음식조혈 CyPA 여 CD147수평교동시기대조조균명현승고(P <0.05),우이모형조위심。제대조조,기여3조균가견내막증생,모형조환가견전형 AS반괴형성;반괴내급증생내막균가견 CyPA 급 CD147적표체。모형조、액담동상조、고지음식조최소관강직경급관강면적교대조조균명현감소(P <0.05),내막면적、중막면적、최대내막후도、관강협착도교대조조명현증대(P <0.05)。결론 CyPA 여 CD147재 AS 토혈관중구중적표체명현승고。
Objective To explore the expressions of Cyclophilin A( CyPA)/ CD147 in vascular remodeling of ath-erosclerosis(AS)rabbits. Methods 32 healthy male New Zealand rabbits(3 months)were randomly divided into the control group,model group,cold-induced injury group,hyperlipid group(8 in each group). Expressions of CyPA and CD147 in serum of rabbits were tested at the beginning and the 13th week,respectively. All rabbits were sacrificed after 13 weeks. Then paraffin-embedded sections were used for HE staining and special staining for elas-tic fiber to analyze the changes of vascular morphology. Additionally,macrophagocyte / CyPA/ CD147 monoclonal antibodies were applied to immunohistochemistical staining and the changes of related vascular remodeling indexes after arterial injury were observed using computerized image analyzing system. Results The expressions of CyPA and CD147 in serum increased significantly in the model group,cold-induced injury group and hyperlipid group compared with those in control group at 13th week(P < 0. 05),respectively,especially the model group. Hyper-plastic intima was found in the model group,cold-induced injury group and hyperlipid group,but not in the control group. Additionally,atherosclerotic plaque could be found in the model group. There were high expressions of CyPA and CD147 in atherosclerotic plaques and hyperplastic intima in the model group,also in the cold-induced in-jury group and hyperlipid group. The minimum lumen diameter and lumen area significantly decreased in these three group compared with the control group(P < 0. 05). However,the intima area,media area,maximum intima thickness and lumen stenosis increased significantly( P < 0. 05). Conclusion High expressions of CyPA and CD147 are found in Remodeling Vascular of atherosclerosis rabbits.
