探讨慢病毒介导的靶向 VEGF 小干扰 RNA 联合应用化疗药物5-FU 诱导人乳腺癌细胞 MCF-7凋亡的机制。以携带 VEGF siRNA 的慢病毒载体感染 MCF-7细胞,应用 RT-PCR 和 Western blot 分别检测各组VEGF mRNA、VEGF 蛋白及凋亡相关蛋白的表达,流式细胞术检测细胞凋亡。结果表明,慢病毒 VEGF siRNA干扰组细胞 VEGF mRNA 和蛋白表达水平明显低于对照组,凋亡相关蛋白 P53及 P21表达上调,而 SIRT1、Bcl-2及 Survivin 表达下调。流式细胞术检测显示慢病毒干扰组及5-FU 组细胞凋亡率显著升高,联合治疗组的协同作用更为明显。上述结果表明:慢病毒介导的 RNA 干扰能明显抑制 MCF-7细胞 VEGF 的表达,通过下调SIRTI 蛋白的表达,导致 P53蛋白表达上调,并调控其下游 P21、bcl-2和 Survivin 的表达,从而诱导 MCF-7细胞的凋亡,并且提高了 MCF-7对5-FU 的敏感性。
探討慢病毒介導的靶嚮 VEGF 小榦擾 RNA 聯閤應用化療藥物5-FU 誘導人乳腺癌細胞 MCF-7凋亡的機製。以攜帶 VEGF siRNA 的慢病毒載體感染 MCF-7細胞,應用 RT-PCR 和 Western blot 分彆檢測各組VEGF mRNA、VEGF 蛋白及凋亡相關蛋白的錶達,流式細胞術檢測細胞凋亡。結果錶明,慢病毒 VEGF siRNA榦擾組細胞 VEGF mRNA 和蛋白錶達水平明顯低于對照組,凋亡相關蛋白 P53及 P21錶達上調,而 SIRT1、Bcl-2及 Survivin 錶達下調。流式細胞術檢測顯示慢病毒榦擾組及5-FU 組細胞凋亡率顯著升高,聯閤治療組的協同作用更為明顯。上述結果錶明:慢病毒介導的 RNA 榦擾能明顯抑製 MCF-7細胞 VEGF 的錶達,通過下調SIRTI 蛋白的錶達,導緻 P53蛋白錶達上調,併調控其下遊 P21、bcl-2和 Survivin 的錶達,從而誘導 MCF-7細胞的凋亡,併且提高瞭 MCF-7對5-FU 的敏感性。

The mechanism of the apoptosis of human breast cancer cell line MCF-7 induced by lentivirus with media-tion of target-oriented VEGF siRNA combined with the anticancer drug 5-FU was investigated. Lentiviral vectors carry-ing VEGF siRNA was infected into human breast cancer MCF-7 cells,the level of VEGF mRNA,VEGF protein and apoptosis related proteins of each group were measured by RT-PCR and Western blot respectively,cell apoptosis was analyzed by flow cytometry. The results showed that in the lentivirus VEGF siRNA interference group the VEGF mR-NA and protein expression level was significantly lower than that in the control group,the expression of the apoptosis related proteins P53 and P21 increased,while SIRT1,Bcl-2 and Survivin decreased. Flow cytometry results showed that the cell apoptosis rate in the interference group and 5-FU group were increased significantly,the synergistic effect of the combined treatment group was more obvious. The above-mentioned results indicated that lentivirus mediated siRNA interfering could significantly suppressed VEGF expression in MCF-7 cell,induced the apoptosis of MCF-7 cells by down-regulating the expression of SIRTl,and induced up-regulating the expression of P53,which regulated the expression of its down stream P21,Bcl-2 and Survivin,and induced the apoptosis of MCF-7,moreover improved the sensitivity of MCF-7 to 5-FU.
