以大蒜根尖为材料,比较不同处理方案下的实验效果,找到植物细胞有丝分裂实验的最佳方案。实验结果显示:观察大蒜根尖有丝分裂最佳培养温度为4℃,分裂指数达40.70%;最佳培养时间为3d,分裂指数达29.00%;10∶00~11∶00为最适取材时间,分裂指数为29.00%.此外,当解离液中浓盐酸与95%乙醇体积比为4∶1、染液为1g/L的番红时染色效果较好,能清晰地显示有丝分裂相。

A variety of Pretreatment regimens were set with the garlic root tip.Comparing the experimental results under dif-ferent treatments were to find the best scheme of the plant cell mitosis experiment.The results show that:the best culture tem-perature for observing the plant cell mitosis is 4℃, with the mitotic index up to 40.70%;the best culture time is 3 days, with the mitotic index up to 29.00%;the most optimal time for sampling is 10∶00-11∶00, with the mitotic index up to 29.00%. In addition, the mitotic characters could be observed clearly when the volume ratio of hydrochloric acid and alcohol in the dis-sociation solution is 4∶1 and the dye is 1g/L of safranin.
