目的:探讨DNA错配修复蛋白MLH1及原癌基因HER-2在散发性结直肠癌中的表达及相关性研究。方法对150例散发性结直肠癌分别进行免疫组化MHL1及原位杂交HER-2检测,观察两种基因的阳性率、与临床特征的相关性。结果免疫组化显示MHL1有47例阳性,阳性率为31.3%,与患者的年龄、肿瘤分化程度及脉管侵犯之间有统计学差异(P<0.05),原位杂交显示HER-2有38例阳性,阳性率为25.3%,并且与肿瘤大小、分化程度及脉管侵犯之间有统计学差异(P<0.05)。MHL1与HER-2在结直肠癌中的表达无显著性相关(P>0.05)。结论 MHL1与HER-2分别独立作用于结直肠癌形成机制,联合检测MHL1及HER-2,可以作为肿瘤预后及靶向治疗的参考指标。

목적:탐토DNA착배수복단백MLH1급원암기인HER-2재산발성결직장암중적표체급상관성연구。방법대150례산발성결직장암분별진행면역조화MHL1급원위잡교HER-2검측,관찰량충기인적양성솔、여림상특정적상관성。결과면역조화현시MHL1유47례양성,양성솔위31.3%,여환자적년령、종류분화정도급맥관침범지간유통계학차이(P<0.05),원위잡교현시HER-2유38례양성,양성솔위25.3%,병차여종류대소、분화정도급맥관침범지간유통계학차이(P<0.05)。MHL1여HER-2재결직장암중적표체무현저성상관(P>0.05)。결론 MHL1여HER-2분별독립작용우결직장암형성궤제,연합검측MHL1급HER-2,가이작위종류예후급파향치료적삼고지표。
Objective To explore the DNA mismatch repair proteins MLH1 and proto-oncogenes HER-2 expression and correlation studies in sporadic colorectal cancer. Methods Based on 150 cases of sporadic colorectal cancer were MHL1 immunohistochemistry and in situ hybridization detection of HER-2, to observe the correlation between the clinical features, with the positive rate of two gene. Results Immunohistochemistry showed that MHL1 was positive in 47 cases, the positive rate was 31.3%,and there is statistical difference with tumor size, differentiation and vascular invasion (P<0.05). In situ hybridization showed that HER-2 was positive in 38 cases, the positive rate was 25.3%, and there is statistical difference with tumor size, differentiation and vascular invasion (P<0.05). Expression of MHL1 and HER-2 in colorectal cancer was not significantly related (P>0.05). Conclusion MHL1 and HER-2 independent role in colorectal cancer formation mechanism, respectively, The combined detection of MHL1 and HER-2, can be used as a reference index of tumor prognosis and targeted therapy.
