目的:观察不同治法对骨质疏松症小鼠骨骼、骨骼肌Ihh含量变化的影响,探讨中医防治骨质疏松症的作用机制。方法除正常组外,将OPG基因敲除小鼠随机分为5组,即模型对照组(模型组)、补肾中药组(补肾组)、健脾中药组(健脾组)、活血化瘀中药组(活血组)和对照药组(福善美组)。采用酶联免疫法测定小鼠骨骼、骨骼肌Ihh含量变化。结果①与正常组比较,模型组小鼠骨骼肌的Ihh含量显著降低( P<0.01);与模型组比较,补肾组小鼠骨骼肌的Ihh含量明显升高( P<0.05),其次为健脾组(P<0.05)。②与正常组比较,模型组小鼠骨骼的Ihh含量显著降低(P<0.01);与模型组比较,补肾组小鼠骨骼肌的Ihh含量明显升高(P<0.01),其次为健脾组(P<0.01),活血组和福善美组升高程度也比较明显(P<0.01)。结论①骨质疏松症的形成,可能与骨骼、骨骼肌Ihh含量异常变化有关。②补肾、健脾方法通过提高骨质疏松症大鼠的骨骼、骨骼肌Ihh的含量,对骨质疏松症具有一定的防治作用。

목적:관찰불동치법대골질소송증소서골격、골격기Ihh함량변화적영향,탐토중의방치골질소송증적작용궤제。방법제정상조외,장OPG기인고제소서수궤분위5조,즉모형대조조(모형조)、보신중약조(보신조)、건비중약조(건비조)、활혈화어중약조(활혈조)화대조약조(복선미조)。채용매련면역법측정소서골격、골격기Ihh함량변화。결과①여정상조비교,모형조소서골격기적Ihh함량현저강저( P<0.01);여모형조비교,보신조소서골격기적Ihh함량명현승고( P<0.05),기차위건비조(P<0.05)。②여정상조비교,모형조소서골격적Ihh함량현저강저(P<0.01);여모형조비교,보신조소서골격기적Ihh함량명현승고(P<0.01),기차위건비조(P<0.01),활혈조화복선미조승고정도야비교명현(P<0.01)。결론①골질소송증적형성,가능여골격、골격기Ihh함량이상변화유관。②보신、건비방법통과제고골질소송증대서적골격、골격기Ihh적함량,대골질소송증구유일정적방치작용。
Objective To approach the mechanism of different therapies in the treatment of osteoporosis by investigating Ihh expression in the bone and skeletal muscle in mice.Methods Except in the WT group, OPG knockout mice were divided into osteoporosis model group, invigorating kidney group, invigorating spleen group, promoting blood flow group, and Fosamax group. Enzyme-linked immunosorbent assay were performed to analyze the content of Ihh in the bone and skeletal muscle.Results (1) All the OPG-/-mice showed significantly lower Ihh expression in the skeletal muscle than mice in WT group.Ihh expression in invigorating kidney group increased significantly than in invigorating spleen group and in model control ( P <0.05 ) .( 2 ) Ihh expression in bone in the model group decreased significantly comparing to that in WT group (P<0.01).Comparing to that in model group, Ihh expression increased the highest in invigorating kidney group (P<0.01), followed by that in invigorating spleen group.It also increased in promoting blood flow group and Fosamax group.Conclusion ( 1 ) Osteoporosis is related to the change of Ihh expression in bone and skeletal muscle.( 2 ) Treatment with invigorating the kidney and invigorating spleen shows effective on osteoporosis probably by up-regulating Ihh expression in the bone and skeletal muscle.
