本试验旨在研究戊酸雌二醇对雌性巴马香猪脂质代谢的影响及其作用机制。采用单因子试验设计,选取20头健康的35日龄雌性巴马香猪随机分为2组,即对照组与试验组,每组10个重复,每个重复1头猪。对照组饲喂基础饲粮,试验组饲喂在基础饲粮中添加0.66 mg/kg戊酸雌二醇(相当于0.5 mg/kg雌二醇)的试验饲粮,试验周期为120 d。结果表明:1)饲粮添加戊酸雌二醇显著降低了雌性巴马香猪末体重、体增重及体脂肪率( P<0.05)。2)饲粮添加戊酸雌二醇显著降低了雌性巴马香猪血液葡萄糖、甘油三酯及低密度脂蛋白含量( P<0.05)。3)与对照组相比,试验组雌性香猪背部皮下脂肪中脂肪酸合成酶( FAS)、乙酰辅酶-A羧化酶( ACC)及脂蛋白脂肪酶(LPL)的mRNA相对表达水平显著降低(P<0.05),激素敏感脂肪酶(HSL)、脂肪甘油三酯脂酶(ATGL)及雌激素受体-β(ER-β)的mRNA相对表达水平显著升高(P<0.05),过氧化物酶体增殖因子受体-γ( PPAR-γ)、过氧化物酶体增殖因子受体-α( PPAR-α)及雌激素受体-α(ER-α)的mRNA相对表达水平无显著差异(P>0.05);腹部皮下脂肪中的FAS、ACC及LPL的mRNA相对表达水平显著降低(P<0.05),PPAR-α、HSL、ATGL、ER-α及ER-β的mRNA相对表达水平显著升高( P<0.05),PPAR-γ的mRNA相对表达水平无显著差异( P>0.05);肾周脂肪中的PPAR-γ、FAS、ER-α及ER-β的mRNA相对表达水平显著降低(P<0.05),HSL、ATGL及LPL的mRNA相对表达水平显著升高( P<0.05),ACC及PPAR-α的mRNA相对表达水平无显著差异( P>0.05)。由此可见,戊酸雌二醇可以有效地控制雌性巴马香猪的体增重及脂肪沉积,并且能有效地改善血糖及血脂水平。戊酸雌二醇可能通过2个途径来调节雌性香猪白色脂肪组织中的脂质代谢:其一,通过对脂肪合成及脂肪分解相关基因表达的调节来影响脂质代谢;其二,通过对雌激素受体基因表达的调节来调控LPL的表达,进而影响脂肪的合成代谢。
本試驗旨在研究戊痠雌二醇對雌性巴馬香豬脂質代謝的影響及其作用機製。採用單因子試驗設計,選取20頭健康的35日齡雌性巴馬香豬隨機分為2組,即對照組與試驗組,每組10箇重複,每箇重複1頭豬。對照組飼餵基礎飼糧,試驗組飼餵在基礎飼糧中添加0.66 mg/kg戊痠雌二醇(相噹于0.5 mg/kg雌二醇)的試驗飼糧,試驗週期為120 d。結果錶明:1)飼糧添加戊痠雌二醇顯著降低瞭雌性巴馬香豬末體重、體增重及體脂肪率( P<0.05)。2)飼糧添加戊痠雌二醇顯著降低瞭雌性巴馬香豬血液葡萄糖、甘油三酯及低密度脂蛋白含量( P<0.05)。3)與對照組相比,試驗組雌性香豬揹部皮下脂肪中脂肪痠閤成酶( FAS)、乙酰輔酶-A羧化酶( ACC)及脂蛋白脂肪酶(LPL)的mRNA相對錶達水平顯著降低(P<0.05),激素敏感脂肪酶(HSL)、脂肪甘油三酯脂酶(ATGL)及雌激素受體-β(ER-β)的mRNA相對錶達水平顯著升高(P<0.05),過氧化物酶體增殖因子受體-γ( PPAR-γ)、過氧化物酶體增殖因子受體-α( PPAR-α)及雌激素受體-α(ER-α)的mRNA相對錶達水平無顯著差異(P>0.05);腹部皮下脂肪中的FAS、ACC及LPL的mRNA相對錶達水平顯著降低(P<0.05),PPAR-α、HSL、ATGL、ER-α及ER-β的mRNA相對錶達水平顯著升高( P<0.05),PPAR-γ的mRNA相對錶達水平無顯著差異( P>0.05);腎週脂肪中的PPAR-γ、FAS、ER-α及ER-β的mRNA相對錶達水平顯著降低(P<0.05),HSL、ATGL及LPL的mRNA相對錶達水平顯著升高( P<0.05),ACC及PPAR-α的mRNA相對錶達水平無顯著差異( P>0.05)。由此可見,戊痠雌二醇可以有效地控製雌性巴馬香豬的體增重及脂肪沉積,併且能有效地改善血糖及血脂水平。戊痠雌二醇可能通過2箇途徑來調節雌性香豬白色脂肪組織中的脂質代謝:其一,通過對脂肪閤成及脂肪分解相關基因錶達的調節來影響脂質代謝;其二,通過對雌激素受體基因錶達的調節來調控LPL的錶達,進而影響脂肪的閤成代謝。

To investigate the effect of estradiol valerate on lipid metabolism of female Bama Xiang-pigs and its mechanism, a total of 20 healthy female Bama Xiang-pigs of 35 days old were selected and randomly divided into two groups ( control group and experimental group) with each group had 10 replicates and each replicate had 1 pig. The pigs in the control group were fed a basal diet, while the pigs in the experimental group were fed the basal diet supplemented with 0. 66 mg/kg estradiol valerate ( equivalent to 0. 5 mg/kg estradiol) for 120 d. The results showed as follows:1) diets supplemented with estradiol valerate significantly decreased the final body weight, body weight gain and body fat percentage of female Bama Xiang-pigs ( P<0.05) . 2) Diets supplemented with estradiol valerate significantly reduced the contents of glucose ( GLU) , triglyceride ( TG) and low density lipoprotein ( LDL) in blood of female Bama Xiang-pigs ( P<0.05) . 3) Compared with control group, the mRNA relative expression levels of fatty acid synthetase ( FAS) , acetyl CoA carboxylase ( ACC) and lipoprteinlipase ( LPL) in dorsal subcutaneous adipose of female Bama Xiang-pigs in experimental group were significantly decreased ( P<0.05) , and the mRNA relative expression levels of hormone-sensitive lipase ( HSL) , adipose triglyceride lipase ( ATGL) and estrogen receptor-β ( ER-β) were significantly increased ( P<0.05) , while the mRNA relative expression levels of peroxisome proliferators-activated receptor-γ( PPAR-γ) , peroxisome proliferators-activated receptor-α ( PPAR-α) and estrogen receptor-α ( ER-α) were not significant-ly different ( P>0.05);the mRNA relative expression levels of FAS, ACC and LPL in abdominal subcutaneous adipose were significantly decreased ( P<0.05) , and the mRNA relative expression levels of PPAR-α, HSL, ATGL, ER-α and ER-β were significantly increased ( P<0.05) , while the mRNA relative expression levels of PPAR-γ was not significantly different (P>0.05); the mRNA relative expression levels of PPAR-γ, FAS, ER-α and ER-βin perirenal adipose were significantly decreased ( P<0.05) , and the mRNA relative expression levels of HSL, ATGL and LPL were significantly increased (P<0.05), while the mRNA relative expression levels of ACC and PPAR-α were not significantly different ( P>0.05) . Those results indicate that estradiol va-lerate can control the body weight gain and fat deposition and improve the levels of blood GLU and TG of fe-male Bama Xiang-pigs effectively. In addition, there may be exist two ways for estradiol valerate to regulate the lipid metabolism, one is that influencing the lipid metabolism in different white adipose tissues by regulating the expression of genes related to lipogenic and lipolytic metabolism, the other one is that modulating the ex-pression of LPL by regulating the expression of estrogen receptors, then to affect the lipogenic metabolism.
