目的:探讨硇洲马尾藻褐藻多酚对去卵巢合并高血脂小鼠骨丢失的保护作用。方法雌性昆明小鼠80只,18-22g,4周龄,随机分成8组,分别为正常组、假手术组、去卵巢组、去卵巢+P-407组、去卵巢+P-407+辛伐他汀组、去卵巢+P-407+褐藻多酚低浓度组、去卵巢+P-407+褐藻多酚中浓度组和去卵巢+P-407+褐藻多酚高浓度组。除正常组、假手术组和去卵巢组外,其余各组每隔3天腹腔注射P-407(560 mg/kg)。褐藻多酚组每天灌胃,浓度分别为125 mg/kg、250 mg/kg和500 mg/kg。实验进行90天后,经眼眶采血收集血清及两侧股骨。结果各浓度褐藻多酚组小鼠血脂指标(胆固醇、甘油三脂、低密度脂蛋白)均比去卵巢+P-407组有明显降低( P<0.05),且与给药浓度呈正相关;对甘油三酯的消除效果优于辛伐他汀( P<0.05),与正常组相近(P>0.05)。去卵巢+P-407组小鼠血清中丙二醛(MDA)含量明显高于其他组(P<0.05),超氧化物歧化酶( SOD)含量则明显低于其他各组( P<0.05)。而各浓度治疗组小鼠血清中SOD和MDA含量与对照组相比未见显著性差异(P>0.05)。去卵巢+P-407组股骨生物力学指标与正常组相比有明显的下降(P<0.05);各治疗组股骨最大负载值、骨刚性系数值、断裂负荷值、断裂负荷值曲线下面积值和最大负荷值曲线下面积值均比模型组有所提高,其中断裂负荷值曲线下面积值比模型组明显升高( P<0.05)。结论硇洲马尾藻多酚通过抑制氧化应激预防去卵巢合并高血脂小鼠的骨丢失。

목적:탐토뇨주마미조갈조다분대거란소합병고혈지소서골주실적보호작용。방법자성곤명소서80지,18-22g,4주령,수궤분성8조,분별위정상조、가수술조、거란소조、거란소+P-407조、거란소+P-407+신벌타정조、거란소+P-407+갈조다분저농도조、거란소+P-407+갈조다분중농도조화거란소+P-407+갈조다분고농도조。제정상조、가수술조화거란소조외,기여각조매격3천복강주사P-407(560 mg/kg)。갈조다분조매천관위,농도분별위125 mg/kg、250 mg/kg화500 mg/kg。실험진행90천후,경안광채혈수집혈청급량측고골。결과각농도갈조다분조소서혈지지표(담고순、감유삼지、저밀도지단백)균비거란소+P-407조유명현강저( P<0.05),차여급약농도정정상관;대감유삼지적소제효과우우신벌타정( P<0.05),여정상조상근(P>0.05)。거란소+P-407조소서혈청중병이철(MDA)함량명현고우기타조(P<0.05),초양화물기화매( SOD)함량칙명현저우기타각조( P<0.05)。이각농도치료조소서혈청중SOD화MDA함량여대조조상비미견현저성차이(P>0.05)。거란소+P-407조고골생물역학지표여정상조상비유명현적하강(P<0.05);각치료조고골최대부재치、골강성계수치、단렬부하치、단렬부하치곡선하면적치화최대부하치곡선하면적치균비모형조유소제고,기중단렬부하치곡선하면적치비모형조명현승고( P<0.05)。결론뇨주마미조다분통과억제양화응격예방거란소합병고혈지소서적골주실。
Objective To investigate the protective effect of phlorotannins from Sargassum naozhouense on bone loss in ovariectomized mice with hyperlipidemia.Methods Eighty female 4-week-old Kunming mice (20-22 g) were randomly divided into 8 groups:Control, SHAM, OVX, OVX+P-407, OVX+P-407+S, OVX+P-407+LP, OVX+P-407+MP, and OVX+P-407+HP group.Mice in each group were given P-407 560mg/kg, i.p.every 3 days except in Control, SHAM, and OVX group. Phlorotannins of 125, 250, and 500mg/kg were given i.g.every day, respectively.The mice were sacrificed ninety days later, and the serum and bilateral femurs were collected.Results The concentrations of cholesterol, triglyceride, and low density lipoprotein in mice of remedy groups were lower than those in OVX +P-407 group ( P <0.05 ) , and it was positively correlated with the concentration of phlorotannins.The decrease rate of triglyceride in phlorotannins treated groups was better than that in OVX+P-407+S group ( P<0.05) .The concentration of malondialdehyde ( MDA) in mice of OVX+P-407 group was higher than that in the other groups (P<0.05), but the superoxide dismutase (SOD) concentration in OVX+P-407 group was lower than that in the other groups (P<0.05).Both SOD and MDA concentration of the remedy groups were close to the blank control group (P>0.05). Biomechanical indexes in the femurs of OVX+P-407 group was significantly lower than those in the blank control group ( P<0. 05).The values of maximum load, rigidity coefficient, breaking load, the area under the maximum load curve, and breaking load curve in the remedy groups were higher than those in OVX+P-407 group.Among these indexes, the value of breaking load curve was significantly higher ( P <0.05 ) . Conclusion Phlorotannins from sargassum naozhouense can protect bone loss in ovariectomized mice with hyperlipidemia by inhibiting oxidative stress in vivo.
