目的:观察少突胶质前体细胞(OPCs)移植对实验性变态反应性脑脊髓炎(EAE)大鼠脊髓有髓神经纤维髓鞘形态的影响。方法选择30只健康大鼠随机分为正常组、模型对照组、治疗组,每组10只。正常组正常饲养,模型对照组、治疗组采用大鼠脊髓匀浆与完全弗氏佐剂混合做抗原单点皮下注射造模,造模后1周治疗组由尾静脉注入OPCs悬液,模型对照组注入同等剂量生理盐水,连续测量大鼠体重的改变,将各组大鼠心脏灌注处死,取脊髓制成切片,用髓鞘碱性蛋白(MBP)免疫组化染色并观察。结果造模后1周模型对照组与治疗组大鼠体重均下降。移植后,治疗组大鼠体重下降有所缓解;与正常组比较,模型对照组大鼠运动功能评分降低,差异有统计学意义(P<0.05);与模型对照组比较,治疗组运动评分升高,差异有统计学意义(P<0.05);与模型对照组比较,治疗组MBP免疫组化阳性的髓鞘数量增多。结论OPCs移植可促进脱髓鞘模型大鼠运动功能和脊髓髓鞘的修复。

목적:관찰소돌효질전체세포(OPCs)이식대실험성변태반응성뇌척수염(EAE)대서척수유수신경섬유수초형태적영향。방법선택30지건강대서수궤분위정상조、모형대조조、치료조,매조10지。정상조정상사양,모형대조조、치료조채용대서척수균장여완전불씨좌제혼합주항원단점피하주사조모,조모후1주치료조유미정맥주입OPCs현액,모형대조조주입동등제량생리염수,련속측량대서체중적개변,장각조대서심장관주처사,취척수제성절편,용수초감성단백(MBP)면역조화염색병관찰。결과조모후1주모형대조조여치료조대서체중균하강。이식후,치료조대서체중하강유소완해;여정상조비교,모형대조조대서운동공능평분강저,차이유통계학의의(P<0.05);여모형대조조비교,치료조운동평분승고,차이유통계학의의(P<0.05);여모형대조조비교,치료조MBP면역조화양성적수초수량증다。결론OPCs이식가촉진탈수초모형대서운동공능화척수수초적수복。
Objective To observe the influence of oligodendrocyte precursor cells transplantation to motor function and myelinated nerve fiber morphology of experimental allergic encephalomyelitis rats. Methods 30 healthy rats were ran-domly divided into normal group,model control group,treatment group.Normal group was given normal breeding,spinal cord homogenate and complete Freund’s adjuvant were mixed and injected into rats in the model control group and treatment group.About 1 week later,the treatment group rats were given intravenous injection of oligodendrocyte precur-sor cells,model control group was injected equal dose of normal saline,the change of weight in rats were measured con-tinuously.Rats in each group were killed by cardiac perfusion,spinal cord was made into slices,MBP immunohistochemi-cal staining was given and observed. Results Compared with the normal group,the weight of model control group de-creased after one week for making model.After transplantation,compared with the model control group,the weight of treatment group increased.Compared with the normal group,clinical score of model control group decreased,the differ-ence had statistically significance (P<0.05).Comparing with the control group,clinical score of treatment group in-creased,the difference had statistically significance (P<0.05).Comparing with the control group,the number of myelin which was positive in MBP immunohistochemistry test in model control group increased. Conclusion The oligodendro-cyte precursor cells transplantation has certain effect on repairing the motor function and myelin sheath of the myeli-nated nerve fibers in spinal cord.
