目的:检测 miR-20b 在肾癌及癌旁组织中的表达水平,分析 miR-20b 的表达与临床病理特征之间的关系,探究其临床意义。方法提取42例手术切除的肾癌及配对癌旁组织标本中的总RNA,经逆转录获得 cDNA,通过实时荧光定量 PCR 检测研究标本中 miR-20b 的表达量,使用统计学软件分析 miR-20b 表达的差异及表达水平与患者临床病理特征之间的相关性。结果 miR-20b 在肾癌组织中的表达水平明显低于配对癌旁组织(P <0.05),其中有29例(69.048%)肾癌标本 miR-20b表达下调。肾癌组织中 miR-20b 表达量与患者性别、年龄、肾癌组织类型、AJCC 临床分期、TNM 分期无明显相关(P >0.05)。结论 miR-20b 在肾癌组织中的表达明显下调,提示其可能和肾癌的发生发展相关。

목적:검측 miR-20b 재신암급암방조직중적표체수평,분석 miR-20b 적표체여림상병리특정지간적관계,탐구기림상의의。방법제취42례수술절제적신암급배대암방조직표본중적총RNA,경역전록획득 cDNA,통과실시형광정량 PCR 검측연구표본중 miR-20b 적표체량,사용통계학연건분석 miR-20b 표체적차이급표체수평여환자림상병리특정지간적상관성。결과 miR-20b 재신암조직중적표체수평명현저우배대암방조직(P <0.05),기중유29례(69.048%)신암표본 miR-20b표체하조。신암조직중 miR-20b 표체량여환자성별、년령、신암조직류형、AJCC 림상분기、TNM 분기무명현상관(P >0.05)。결론 miR-20b 재신암조직중적표체명현하조,제시기가능화신암적발생발전상관。
Objective To detect the expression level of miR-20b in the renal cell carcinoma (RCC) and paired normal tissues,and analyze the relationships between the expression level of miR-20b and clini-copathologic.Then explore its clinical significance. Methods Total RNA was isolated from 42 RCC tissues and paired adjacent normal tissues resected from nephrectomy ,and cDNA was synthetized by performing reverse transcription.Then real time quantitative PCR(RT-qPCR)was used to detect the expression of miR-20b.We analyzed the relationships between the expression of miR-20b and clinico-pathologic characteristics in tumor and the paired characteristics tissues. Results The expression of miR-20b in RCC tissues was significantly lower than paired adjacent normal tissues(P <0.05 ), and 29(60.048%)RCC tissues showed down-regulation in all tissues which had detectable miR-20b. The results showed that there were no relationships between the expression of miR-20b and patients'gender,age,histology,TNM stage and AJCC stage. Conclusions miR-20b was down-regulated in RCC tissue,and this indicated that miR-20b might be involved in the tumorigenesis and development of RCC.
