Rhizoma Anemarrhenae%Diabetes%Cognitive dysfunction%APP%BACE1
目的:观察知母水煎液对链脲佐菌素( STZ)诱发的糖尿病小鼠模型脑内β-淀粉样蛋白前体蛋白(APP)、淀粉样前体蛋白β-分泌酶(BACE1)蛋白表达的影响,以揭示知母改善糖尿病模型小鼠认知功能的可能机制。方法:采用腹腔注射STZ法造成BALB/C小鼠糖尿病模型,随机分为模型组、知母高剂量组、知母低剂量组、阳性药盐酸二甲双胍组,以正常小鼠作为对照组。各组连续给药30天后,利用Western blot法检测各组小鼠脑中APP和BACE1蛋白的表达情况。结果:与对照组相比,模型组APP和BACE1蛋白表达明显升高( P<0.05);与模型组相比,给药组APP和BACE1蛋白表达显著下降(P<0.05)。结论:知母水煎液能降低由STZ诱发的糖尿病小鼠脑内APP和BACE1蛋白表达的升高。

목적:관찰지모수전액대련뇨좌균소( STZ)유발적당뇨병소서모형뇌내β-정분양단백전체단백(APP)、정분양전체단백β-분비매(BACE1)단백표체적영향,이게시지모개선당뇨병모형소서인지공능적가능궤제。방법:채용복강주사STZ법조성BALB/C소서당뇨병모형,수궤분위모형조、지모고제량조、지모저제량조、양성약염산이갑쌍고조,이정상소서작위대조조。각조련속급약30천후,이용Western blot법검측각조소서뇌중APP화BACE1단백적표체정황。결과:여대조조상비,모형조APP화BACE1단백표체명현승고( P<0.05);여모형조상비,급약조APP화BACE1단백표체현저하강(P<0.05)。결론:지모수전액능강저유STZ유발적당뇨병소서뇌내APP화BACE1단백표체적승고。
Objective:To investigate the effects of Rhizoma Anemarrhenae water solution on the expressions of amyloid precursor protein (APP) andβ-secreatase (BACE1) in the brain of diabetic mice to reveal the pos-sible mechanism that Rhizoma Anemarrhenae may improve the cognitive function of diabetic mice.Methods:The BALB/C mice were treated with STZ by intraperitoneal injection to induce diabetic models, and they were randomly divided into STZ group, STZ supplement with low dose of Rhizoma Anemarrhenae group, STZ sup-plement with high dose of Rhizoma Anemarrhenae group, STZ supplement with metformin hydrochloride group and normal mice as control group.After giving medicine for 30 days, the expressions of APP and BACE1 were detected by Western blot analysis.Results: Compared with those in the control group, the protein levels of APP and BACE1 were significantly increased in STZ group(P<0.05).In addition, protein expressions of APP and BACE1 decreased by different doses of Rhizoma Anemarrhenae ( P<0 .05 ) .Conclusion: Rhizoma Anemarrhenae could reduce the expressions of APP and BACE1 in STZ mice.
