目的:探讨烟酰胺单核苷酸腺苷酰转移酶1(NMNAT1)对慢性帕金森病(PD)小鼠模型(C57BL/6小鼠)黑质中酪氨酸羟化酶(TH)及半胱氨酸天冬氨酸蛋白水解酶3(caspase-3)表达的影响。方法利用重组慢病毒转染1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)制备慢性PD小鼠模型。依据不同处理方式分为3组:对照组[经立体定位仪向小鼠双侧黑质注射空白对照重组慢病毒(LV-GFP),1周后腹腔注射生理盐水];MPTP组(经立体定位仪向小鼠双侧黑质注射LV-GFP,1周后建立MPTP慢性PD模型);NMNAT1组[经立体定位仪向小鼠双侧黑质注射NMNAT1基因的过表达重组慢病毒(LV-NMNAT1),1周后建立MPTP慢性PD模型]。采用Western blot法检测3组小鼠黑质中TH及caspase-3半定量表达水平。结果 NMNAT1呈高表达的NMNAT1组黑质中TH蛋白表达量较MPTP组高(t=7.985,P<0.001),caspase-3蛋白表达量较MPTP组低(t=8.901,P<0.001)。结论 NMNAT1能增加TH表达发挥一定的神经保护作用,并通过减少caspase-3表达水平参与抗细胞凋亡。

목적:탐토연선알단핵감산선감선전이매1(NMNAT1)대만성파금삼병(PD)소서모형(C57BL/6소서)흑질중락안산간화매(TH)급반광안산천동안산단백수해매3(caspase-3)표체적영향。방법이용중조만병독전염1-갑기-4-분기-1,2,3,6-사경필정(MPTP)제비만성PD소서모형。의거불동처리방식분위3조:대조조[경입체정위의향소서쌍측흑질주사공백대조중조만병독(LV-GFP),1주후복강주사생리염수];MPTP조(경입체정위의향소서쌍측흑질주사LV-GFP,1주후건립MPTP만성PD모형);NMNAT1조[경입체정위의향소서쌍측흑질주사NMNAT1기인적과표체중조만병독(LV-NMNAT1),1주후건립MPTP만성PD모형]。채용Western blot법검측3조소서흑질중TH급caspase-3반정량표체수평。결과 NMNAT1정고표체적NMNAT1조흑질중TH단백표체량교MPTP조고(t=7.985,P<0.001),caspase-3단백표체량교MPTP조저(t=8.901,P<0.001)。결론 NMNAT1능증가TH표체발휘일정적신경보호작용,병통과감소caspase-3표체수평삼여항세포조망。
Aim To investigate the effect of nicotinamide mononucleotide adenylyltransferase (NMNAT1) on the expression of tyrosine hydroxylase (TH) and cysteinyl aspartate speciifc proteinase 3 (caspase-3) in the substantia nigra (SN) of chronic MPTP-induced Parkinson’s disease (PD) Mice.Methods Before a week of inducing PD models, lentiviruses were injected bilaterally into the SN of mice. Mice were divided into three groups: a vehicle group, a MPTP group, a NMNAT1 group. The protein levels of TH and caspase-3 were measured by Western blot.Results Compared with MPTP group, TH expression increased (t=7.985,P<0.001) and caspase-3 expression decreased (t=8.901,P<0.001) in NMNAT1 group. Conclusion The results suggested that NMNAT1 may play an important role in protecting dopaminergic neurons, and this role may be effective to down-regulate the expression of caspase-3.
