目的:观察四氯化碳诱导肝纤维化小鼠牛磺酸处理前后肝损伤、纤维化、氧化应激及炎症反应指标的变化,探讨牛磺酸对小鼠肝纤维化的抑制作用。方法将30只小鼠随机分为空白对照组(腹腔注射橄榄油)、模型组和牛磺酸处理组各10只,后两组腹腔注射四氯化碳诱导小鼠肝纤维化,牛磺酸处理组造模的同时在饮水中添加牛磺酸。第10周注射结束后,检测各组小鼠血清透明质酸( HA)、层粘连蛋白( LN)、ALT、AST、IL-2、IL-6及肝脏组织中超氧化物歧化酶( SOD)和丙二醛( MDA);计算肝脏指数;HE和Masson染色法观察肝脏组织病理学变化。结果与模型组比较,牛磺酸处理组小鼠血清中HA、LN、ALT、AST、IL-2及IL-6水平降低(P均<0.05),肝脏组织中SOD水平升高、MDA水平降低,肝脏指数降低(P均<0.05)。病理检查结果显示,牛磺酸处理组肝脏组织中的炎性浸润和脂肪空泡显著减少,无纤维组织沉积。结论牛磺酸对小鼠肝纤维化具有一定的抑制作用。

목적:관찰사록화탄유도간섬유화소서우광산처리전후간손상、섬유화、양화응격급염증반응지표적변화,탐토우광산대소서간섬유화적억제작용。방법장30지소서수궤분위공백대조조(복강주사감람유)、모형조화우광산처리조각10지,후량조복강주사사록화탄유도소서간섬유화,우광산처리조조모적동시재음수중첨가우광산。제10주주사결속후,검측각조소서혈청투명질산( HA)、층점련단백( LN)、ALT、AST、IL-2、IL-6급간장조직중초양화물기화매( SOD)화병이철( MDA);계산간장지수;HE화Masson염색법관찰간장조직병이학변화。결과여모형조비교,우광산처리조소서혈청중HA、LN、ALT、AST、IL-2급IL-6수평강저(P균<0.05),간장조직중SOD수평승고、MDA수평강저,간장지수강저(P균<0.05)。병리검사결과현시,우광산처리조간장조직중적염성침윤화지방공포현저감소,무섬유조직침적。결론우광산대소서간섬유화구유일정적억제작용。
Objective To observe the changes of liver injury, fibrosis, oxidative stress and inflammation indexes be-fore and after treatment of taurine in carbon tetrachloride-induced liver fibrosis of mice, and to investigate the inhibitory effect of taurine on liver fibrosis in mice.Methods Thirty mice were randomly divided into three groups including blank control group, model group and taurine treatment group, 10 mice in each group.Mice in the model and taurine treatment groups were injected carbon tetrachloride in enterocoelia to build the liver fibrosis model.At the same time, mice in the taurine treatment group were fed by taurine in drinking water.After 10 weeks, peripheral blood and serum were collected. Hyaluronic acid ( HA ) , layer adhesion protein ( LN ) , alanine aminotransferase ( ALT ) , aspartate aminotransferase (AST), interleukin 2 (IL-2) and IL-6 in the serum were detected.The liver index was calculated; the super oxide dis-mutase ( SOD) and malondialdehyde ( MDA) level in the liver tissues was detected;and pathological changes of liver tis-sues were stained by HE and Masson.Results Compared with the model group, HA, LN, ALT, AST, IL-2 and IL-6 lev-els in the serum were decreased in the taurine treatment group (all P<0.05);and SOD in the liver tissue was increased, MDA level, liver indexes were decreased in the liver tissues (all P<0.05).The pathological examination showed that the inflammatory infiltrates in the liver tissue and fat cavitation significantly reduced and fibrous tissue deposition could not be found.Conclusion Taurine has a certain inhibitory effect on carbon tetrachloride induced liver fibrosis in mice.
