目的:观察5‐氟尿嘧啶(5‐Fu)对人绒癌细胞株JAR的体外作用。方法:采用M T T比色分析法,观察20、50、100μg/L 3种剂量的5‐Fu对人绒癌细胞株JA R细胞的生长抑制情况;通过免疫组化法检测3种浓度5‐Fu作用于JA R细胞后增殖细胞核抗原Ki67的表达情况。结果:①5‐Fu能抑制JAR细胞的生长,并在一定时间及浓度范围内,表现出剂量依赖性,72h内5‐Fu浓度在25~100μg/L时抑制作用比较明显;②100μg/L的5‐Fu作用于JA R细胞72h后细胞抑制率达73.19%,作用后增殖细胞核抗原Ki67明显下降。结论:①5‐Fu对人绒癌细胞株JA R细胞的生长具有抑制作用,72h内呈剂量依赖性;②5‐Fu抑制了人绒癌细胞株JAR细胞的增殖,细胞可能被阻滞于G0及G1期。

목적:관찰5‐불뇨밀정(5‐Fu)대인융암세포주JAR적체외작용。방법:채용M T T비색분석법,관찰20、50、100μg/L 3충제량적5‐Fu대인융암세포주JA R세포적생장억제정황;통과면역조화법검측3충농도5‐Fu작용우JA R세포후증식세포핵항원Ki67적표체정황。결과:①5‐Fu능억제JAR세포적생장,병재일정시간급농도범위내,표현출제량의뢰성,72h내5‐Fu농도재25~100μg/L시억제작용비교명현;②100μg/L적5‐Fu작용우JA R세포72h후세포억제솔체73.19%,작용후증식세포핵항원Ki67명현하강。결론:①5‐Fu대인융암세포주JA R세포적생장구유억제작용,72h내정제량의뢰성;②5‐Fu억제료인융암세포주JAR세포적증식,세포가능피조체우G0급G1기。
Objective:To investigate the effects of 5‐FU on cell of human choriocarcinoma cell line JAR . Methods :The inhibitory effects of three different concentration of chemotherapeutical drug 5‐Fu(5‐Fluorouracil) (20 ,50 ,100μg/L) on JAR strain were assessed with methylthiazolyl tetrazolium MTT colorimetric assay .The ex‐pression of nuclear proliferative antigen Ki67 in JAR cells were detected with avidin‐biotin complex method ,immu‐no‐histochemical method .Results :① 5‐Fu inhibited the JAR cell growth in a dose‐dependent manner .This effect was obvious at 25~100μg/L in 72h .② When the JAR cells were exposed to 100μg/L 5‐Fu for 72h ,the expression of Ki67 antigen was declined significantly ,the inhibition rate was 73 .19% .Conclusion:① 5‐Fu inhibited the JAR cell growth in a dose‐dependent manner at 72h .② 5‐Fu inhibited the Proliferation of JAR cell ,which may be inhibi‐ted at the Period of G0 and G1 .
