目的:观察硫酸脑苷酯活化Ⅱ型自然杀伤(natural killer,NK)T 细胞对支气管哮喘(简称哮喘)小鼠Ⅰ型 NKT 细胞数量及活性的影响。方法将24只 BALB/c 小鼠随机分为对照组、哮喘组和硫酸脑苷酯组,每组8只。流式细胞术检测肺Ⅱ型及Ⅰ型 NKT 细胞的数量及分泌细胞因子水平;酶联免疫斑点吸附法检测脾Ⅱ型及Ⅰ型 NKT 细胞分泌细胞因子水平;CCK8试剂盒检测脾Ⅰ型 NKT 细胞的增殖能力。结果硫酸脑苷酯组小鼠肺Ⅱ型 NKT 细胞分泌 IL-4和γ干扰素(IFN-γ)水平明显高于哮喘组(P <0.01),但肺及脾Ⅰ型 NKT 细胞分泌 IL-4和 IFN-γ水平和脾Ⅰ型 NKT 细胞对α半乳糖神经酰胺的增殖反应明显低于哮喘组(P 值均<0.01)。结论硫酸脑苷酯活化Ⅱ型 NKT 细胞可以抑制哮喘小鼠Ⅰ型 NKT 细胞的活性。

목적:관찰류산뇌감지활화Ⅱ형자연살상(natural killer,NK)T 세포대지기관효천(간칭효천)소서Ⅰ형 NKT 세포수량급활성적영향。방법장24지 BALB/c 소서수궤분위대조조、효천조화류산뇌감지조,매조8지。류식세포술검측폐Ⅱ형급Ⅰ형 NKT 세포적수량급분비세포인자수평;매련면역반점흡부법검측비Ⅱ형급Ⅰ형 NKT 세포분비세포인자수평;CCK8시제합검측비Ⅰ형 NKT 세포적증식능력。결과류산뇌감지조소서폐Ⅱ형 NKT 세포분비 IL-4화γ간우소(IFN-γ)수평명현고우효천조(P <0.01),단폐급비Ⅰ형 NKT 세포분비 IL-4화 IFN-γ수평화비Ⅰ형 NKT 세포대α반유당신경선알적증식반응명현저우효천조(P 치균<0.01)。결론류산뇌감지활화Ⅱ형 NKT 세포가이억제효천소서Ⅰ형 NKT 세포적활성。
Objective To investigate the effect of sulfatide-activated type Ⅱ NKT cells on the activity of type Ⅰ NKT cells.Methods Twenty-four BALB/c mice were randomly divided into three groups:a normal control group (n =8),an asthmatic group (n =8)and a sulfatide treatment group (n =8). The percentages of lung type Ⅱ NKT cells and type Ⅰ NKT cells in lung MNCs,the expressions of IL-4 and IFN-γof lung type Ⅱ NKT cells,as well as the expressions of CD69,IL-4 and IFN-γof lung type ⅠNKT cells were detected by flow cytometry.The expressions of cytokines of spleen type Ⅰ NKT cells and type Ⅱ NKT cells were detected by ELISPOT.The proliferation function of type Ⅰ NKT cells was detected by CCK8 kit.Results Compared with asthma group,the percentage and the expression of IL-4 and IFN-γon lung type Ⅱ NKT cells in sulfatide treated group were significantly increased.In addition, compared with asthma group,the function of type Ⅰ NKT cells were inhibited in sulfatide treated group, including lower productions of IL-4 and IFN-γof lung and spleen type Ⅰ NKT cells,reduced proliferation of spleen type Ⅰ NKT cells to α-Galcer.Conclusions Sulfatide-activated type Ⅱ NKT cells can inhibit the activity of type Ⅰ NKT cells in a murine model of asthma.
