目的:探讨 MMP-2和 MMP-9 mRNA 在新疆哈萨克族食管鳞癌及其癌旁组织中的表达及二者的相关性。方法选取食管癌手术离体标本40例,每例各取癌组织和对应的癌旁组织一份,采用逆转录-聚合酶链式反应法(RT-PCR 法)检测 MMP-2和 MMP-9 mRNA 在癌及癌旁组织组中的表达。运用 Western-blot 法检测 MMP-2和 MMP-9在癌及癌旁组织组中的蛋白表达。采用 Pearson 相关分析法分析 MMP-2和 MMP-9 mRNA 在癌组织中表达的相关性。结果MMP-2 mRNA 在癌组织和癌旁组织中的阳性表达率分别为95.0%(38/40)和77.5%(31/40),差异有统计学意义(χ2=5.165,P =0.023);MMP-9 mRNA 在癌组织和癌旁组织中的阳性率分别为62.5%(25/40)和10.0%(4/40),差异有统计学意义(χ2=23.854,P =0.000)。MMP-2 mRNA 的表达与食管鳞癌浸润深度有关联(χ2=5.260,P =0.022),与食管鳞癌淋巴结转移无关联(χ2=0.088,P =0.767)。MMP-9 mRNA的表达与食管鳞癌浸润深度有关(χ2=4.365,P =0.037),与食管鳞癌淋巴结转移无关联(χ2=0.156,P =0.693)。MMP-2 mRNA 与 MMP-9 mRNA 在食管鳞癌组织中的表达呈正相关关系(r =0.327,P =0.039)。结论MMP-2和 MMP-9可能参与了新疆哈萨克族食管鳞状细胞癌的肿瘤发生发展,但在淋巴结转移方面可能不发挥关键作用。

목적:탐토 MMP-2화 MMP-9 mRNA 재신강합살극족식관린암급기암방조직중적표체급이자적상관성。방법선취식관암수술리체표본40례,매례각취암조직화대응적암방조직일빈,채용역전록-취합매련식반응법(RT-PCR 법)검측 MMP-2화 MMP-9 mRNA 재암급암방조직조중적표체。운용 Western-blot 법검측 MMP-2화 MMP-9재암급암방조직조중적단백표체。채용 Pearson 상관분석법분석 MMP-2화 MMP-9 mRNA 재암조직중표체적상관성。결과MMP-2 mRNA 재암조직화암방조직중적양성표체솔분별위95.0%(38/40)화77.5%(31/40),차이유통계학의의(χ2=5.165,P =0.023);MMP-9 mRNA 재암조직화암방조직중적양성솔분별위62.5%(25/40)화10.0%(4/40),차이유통계학의의(χ2=23.854,P =0.000)。MMP-2 mRNA 적표체여식관린암침윤심도유관련(χ2=5.260,P =0.022),여식관린암림파결전이무관련(χ2=0.088,P =0.767)。MMP-9 mRNA적표체여식관린암침윤심도유관(χ2=4.365,P =0.037),여식관린암림파결전이무관련(χ2=0.156,P =0.693)。MMP-2 mRNA 여 MMP-9 mRNA 재식관린암조직중적표체정정상관관계(r =0.327,P =0.039)。결론MMP-2화 MMP-9가능삼여료신강합살극족식관린상세포암적종류발생발전,단재림파결전이방면가능불발휘관건작용。
Objective To study the expression and correlation of MMP-2 and MMP-9 mRNA in esophageal squamous cell carcinoma and adjacent tissues of Kazakh patients in Xinjiang.Methods 40 histopathologi-cally-confirmed esophageal cancer and an equal number of corresponding adjacent tissue biopsies were ana-lyzed for the expression of MMP-2 and MMP-9 mRNA in esophageal squamous cell carcinoma and adjacent tissues with RT-PCR method.Western-blot was used to detect the expression of protein of MMP-9 and MMP-2 in the cancer tissues and corresponding adjacent tissues.The Pearson correlation analysis was used to analyze the correlation between MMP-9 and mRNA MMP-2 expression in cancer tissues.Results Posi-tive rates of MMP-2 mRNA expression in cancer tissues and corresponding adjacent tissues were 95.0%(38/40)and 77.5% (31/40)respectively,and the difference was statistically significant (χ2=5.165,P =0.023);Positive rates of MMP-9 mRNA in cancer tissues and corresponding adjacent tissues were 62.5%(25/ 40)and 10.0% (4/40),which was also statistically significant (χ2 =23.854,P =0.000).The expres-sion of MMP-2 mRNA was correlated with the depth of invasion of esophageal squamous cell carcinoma (χ2 =5.260,P =0.022)but had no association with the lymph node metastasis of esophageal squamous cell carcinoma (χ2 =0.088,P =0.767).The expression of MMP-9 mRNA was correlated with the depth of invasion of esophageal squamous cell carcinoma (χ2 =4.365,P =0.037),but there was no association with lymph node metastasis (χ2 =0.156,P =0.693).The expression of mRNA MMP-9 and mRNA MMP-2 in esophageal squamous cell carcinoma was positively correlated (r =0.327,P =0.039).Conclusion MMP-2 and MMP-9 may be involved in the occurrence and development of esophageal squamous cell carcinoma in Kazakh patients but may not play a key role in the lymph node metastasis.
