目的探讨糖尿病性白内障及老年性白内障晶状体上皮细胞(LECs)的凋亡与 Fas、Bax 基因的关系。方法在白内障超声乳化手术中取糖尿病性及老年性白内障患者的晶状体前囊膜标本45例,应用光镜,电镜观察 LECs 的凋亡情况,应用 RT-PCR 定量检测 Fas 蛋白,Bax 蛋白在白内障 LECs 中的表达。结果两种白内障患者的 LECs 呈透明样变性,细胞内有空泡(糖尿病性白内障 LECS 可见增生重叠),细胞多呈扁平状,大小不均,核固缩,染色质凝聚边缘化。糖尿病性白内障 LECs 除可见凋亡小体之外,上述改变较老年性白内障更为严重。两种白内障 LECs 中均有 Fas、Bax 蛋白表达。Fas 蛋白在糖尿病性白内障 LECs 中的表达平均是老年性白内障的13.34倍,Bax 蛋白在糖尿病性白内障 LECs 中的表达平均是老年性白内障的38.12倍,差异有统计学意义(P <0.05)。结论两种白内障 LECs 均出现凋亡,Fas、Bax 基因在白内障 LECS 凋亡中起重要作用。

목적탐토당뇨병성백내장급노년성백내장정상체상피세포(LECs)적조망여 Fas、Bax 기인적관계。방법재백내장초성유화수술중취당뇨병성급노년성백내장환자적정상체전낭막표본45례,응용광경,전경관찰 LECs 적조망정황,응용 RT-PCR 정량검측 Fas 단백,Bax 단백재백내장 LECs 중적표체。결과량충백내장환자적 LECs 정투명양변성,세포내유공포(당뇨병성백내장 LECS 가견증생중첩),세포다정편평상,대소불균,핵고축,염색질응취변연화。당뇨병성백내장 LECs 제가견조망소체지외,상술개변교노년성백내장경위엄중。량충백내장 LECs 중균유 Fas、Bax 단백표체。Fas 단백재당뇨병성백내장 LECs 중적표체평균시노년성백내장적13.34배,Bax 단백재당뇨병성백내장 LECs 중적표체평균시노년성백내장적38.12배,차이유통계학의의(P <0.05)。결론량충백내장 LECs 균출현조망,Fas、Bax 기인재백내장 LECS 조망중기중요작용。
Objective To explore the relationship between lens epithelial cells (LECs)apoptosis of the dia-betic cataract and senile cataract and the expression of Fas,Bax.Methods Forty-five cases were selected from dia-betic and senile patients'anterior lens capsules specimens,using light and electron microscopic to observe apoptosis of LECs,and applying RT-PCR to quantitatively detect expression of Fas and Bax in cataract LECs.Results The two LECs of cataract patients were hyaline degeneration.Cells had vacuoles.(The diabetes cataract LECS visible prolifera-tion overlap).Cells were mostly flat,uneven size,nuclear pyknosis,marginalization of chromatin condensation.In LECs of diabetic cataract,these changes above of diabetic cataract even more serious than those in senile cataract.Two kinds of cataract in the LECs had both protein expression of Fas and Bax.Fas protein expression in diabetic cataract LECs was 13.34 times as high as senile cataract.Bas protein expression in diabetic cataract LECs was 38.12 times as high as se-nile cataract.and significant differences was found between them(P <0.05).Conclusion The typical apoptosis was observed in two kinds of cataract LECs.Fas and Bax genes play an important role in cataract LECs apoptosis.
