目的探讨多壁碳纳米管(MWCNT)对MC3T3‐E1成骨细胞迁移的影响。方法采用超声法分散 MWCNT ,经扫描电镜、投射电镜和动态光散射仪检测分散效果。采用 MWCNT (100 g/mL)处理MC3T3‐E1成骨细胞,原子力显微镜检测细胞表面形态,划痕实验检测细胞迁移情况,Gradientech Tracking软件分析细胞迁移特性。结果 MWCNT获得较好分散,细胞处理12 h后, MWCNT大量吸附于细胞表面。划痕实验显示对照组愈合率为80%,MWCNT处理组愈合率为45%(P<0.01),其迁移距离、直线距离和迁移速度均显著低于对照组。结论 MWCNT可大量吸附于成骨细胞表面并显著抑制M C3 T 3‐E1成骨细胞迁移。

목적탐토다벽탄납미관(MWCNT)대MC3T3‐E1성골세포천이적영향。방법채용초성법분산 MWCNT ,경소묘전경、투사전경화동태광산사의검측분산효과。채용 MWCNT (100 g/mL)처리MC3T3‐E1성골세포,원자력현미경검측세포표면형태,화흔실험검측세포천이정황,Gradientech Tracking연건분석세포천이특성。결과 MWCNT획득교호분산,세포처리12 h후, MWCNT대량흡부우세포표면。화흔실험현시대조조유합솔위80%,MWCNT처리조유합솔위45%(P<0.01),기천이거리、직선거리화천이속도균현저저우대조조。결론 MWCNT가대량흡부우성골세포표면병현저억제M C3 T 3‐E1성골세포천이。
Objective To assess the influence of multiwalled carbon nanotubes (MWCNTs) on migration of MC3T3‐E1 osteoblasts .Methods Ultrasound was used for MWCNTs .The morphology and size of the dispersed MWCNTs were detected by scanning electron microscopy , transmission electron microscopy and dynamic light scattering (DLS) .MC3T3‐E1 cells were treated with MWNCTs (100 μg/mL) ,and the morphology of MC3T3‐E1 cells were detected with atomic force microscopy .Cell migration was measured with wound healing methods ,and the property of cell migration was analyzed by Gradientech Tracking software .Results MWCNTs were well dispersed , and absorbed to the surface of cells after exposure for 12 h .The wound healing assay showed that the wound healing rate after 12 h was 45% for MWCNTs treated MC3T3‐E1 cells versus 80% for non‐treated controls(P<0 .01) .The distance ,direct distance and cell motility velocity of MC3T3‐E1 cells also reduced similarly due to the presence of MWCNTs .Conclusion MWCNTs absorb to the surface of MC3T3‐E1 cells and significantly inhibit the migration of osteoblasts .
