目的：研究骨保护素（ OPG）是否能够影响人软骨肉瘤细胞的增殖以及其对人软骨肉瘤细胞表达基质金属蛋白酶-13（MMP-13）的影响。方法购买并传代人软骨肉瘤细胞（SW1353），用不同浓度的OPG刺激后，分别采用MTT方法检测其增殖率，ELISA、RT-PCR以及Western-blot方法检测MMP-13的表达水平。结果随着OPG浓度的增加，人软骨肉瘤细胞的增殖明显增加，并且显著降低了MMP-13的表达。结论 OPG可以促进人软骨肉瘤细胞的增殖，减少MMP-13的表达，进而可能产生对软骨细胞的保护作用，有望在临床中应用于骨关节病的防治。
목적：연구골보호소（ OPG）시부능구영향인연골육류세포적증식이급기대인연골육류세포표체기질금속단백매-13（MMP-13）적영향。방법구매병전대인연골육류세포（SW1353），용불동농도적OPG자격후，분별채용MTT방법검측기증식솔，ELISA、RT-PCR이급Western-blot방법검측MMP-13적표체수평。결과수착OPG농도적증가，인연골육류세포적증식명현증가，병차현저강저료MMP-13적표체。결론 OPG가이촉진인연골육류세포적증식，감소MMP-13적표체，진이가능산생대연골세포적보호작용，유망재림상중응용우골관절병적방치。
Objective The purpose of this research was to investigate the potential effect of OPG( osteoprotegerin)upon the inhibition of matrix metalloprotease-13(MMP-13)expression in chondrosarcoma cell matrix. Methods After purchasing and passaging human chondrosar-coma cells(SW1353)which were stimulated with OPG at different concentrations,we adoptted MTT(methylthiazoletetrazolium)method to test the reproduction rate,employed ELISA,RT-PCR and Western-blot to detect the expression level of MMP-13 respectively. Results The re-sult showed that chondrosarcoma cells reproduction was dramatically enhanced and their MMP13 expression was inhibited with an increase in OPG concentration. Conclusion MMP-13 expression in chondrosarcoma cells was inhibited by OPG. And OPG can be used in the prevention of Os-teoarthritis.
