目的:探讨裙带菜多糖在异丙肾上腺素(ISO)诱导的心脏纤维化中的作用。方法将40只C57BL/6J小鼠随机分为对照组、ISO组、ISO+裙带菜多糖组、裙带菜多糖组。ISO组连续皮下注射ISO 14 d(前3 d 10 mg·kg-1·d-1,后11 d 5 mg· kg-1·d-1),ISO+裙带菜多糖组除做上述ISO处理外,ISO处理前7 d开始给予裙带菜多糖200 mg·kg-1·d-1灌胃,持续到ISO皮下注射第14天;裙带菜多糖组连续21 d裙带菜多糖200 mg·kg-1·d-1灌胃;对照组以生理盐水代替ISO皮下注射。心脏超声检测各组小鼠心功能的改变,病理染色检测心脏纤维化程度,实时定量PCR检测转化生长因子-β(TGF-β)、Ⅰ型胶原α(CollagenⅠα)和Ⅲ型胶原(CollagenⅢ)的mRNA表达量的变化,Western Blot检测各组小鼠心脏自噬的改变。结果裙带菜多糖明显改善心功能,减少ISO诱导的心脏纤维化程度,心脏胶原蛋白CollagenⅠα和CollagenⅢ的mRNA表达量比ISO组显著降低(P<0.05),且裙带菜多糖能减少ISO诱导的心脏自噬。结论裙带菜能减轻ISO诱导的心脏纤维化,其主要是通过降低ISO诱导的心脏自噬而发挥作用的。

목적:탐토군대채다당재이병신상선소(ISO)유도적심장섬유화중적작용。방법장40지C57BL/6J소서수궤분위대조조、ISO조、ISO+군대채다당조、군대채다당조。ISO조련속피하주사ISO 14 d(전3 d 10 mg·kg-1·d-1,후11 d 5 mg· kg-1·d-1),ISO+군대채다당조제주상술ISO처리외,ISO처리전7 d개시급여군대채다당200 mg·kg-1·d-1관위,지속도ISO피하주사제14천;군대채다당조련속21 d군대채다당200 mg·kg-1·d-1관위;대조조이생리염수대체ISO피하주사。심장초성검측각조소서심공능적개변,병리염색검측심장섬유화정도,실시정량PCR검측전화생장인자-β(TGF-β)、Ⅰ형효원α(CollagenⅠα)화Ⅲ형효원(CollagenⅢ)적mRNA표체량적변화,Western Blot검측각조소서심장자서적개변。결과군대채다당명현개선심공능,감소ISO유도적심장섬유화정도,심장효원단백CollagenⅠα화CollagenⅢ적mRNA표체량비ISO조현저강저(P<0.05),차군대채다당능감소ISO유도적심장자서。결론군대채능감경ISO유도적심장섬유화,기주요시통과강저ISO유도적심장자서이발휘작용적。
Objective To investigate the effect of wakame polysaccharide on isoproterenol (ISO)-induced cardiac fibrosis.Methods Forty C57BL/6J mice were randomized into vehicle group,ISO group,ISO +wakame polysaccharide group and wakame polysaccharide group.The vehicle group was treated with saline.Cardiac fibrosis was induced by subcutaneous ISO injection daily (10 mg·kg-1 ·d-1 for 3 days and 5 mg·kg-1·d-1 for 11 days),and wakame polysaccharide (200 mg·kg-1·d-1 for 21 days,starting before ISO in-jection)was administered by oral gavage.After 14 days,animal echo-cardiography was used to preliminarily evaluate the success of the cardiac fibrosis model,and then the mice were killed.Results Wakame polysaccharide significantly improved cardiac function.The cardiac fibrosis was reduced in polysaccharide group as assessed by pathological staining and expression levels of TGFβ,CollagenⅠαand CollagenⅢmRNA (P<0.05).Moreover,wakame polysaccharide significantly reduced ISO-induced autophagy in heart.Conclu-sion Wakame polysaccharide can inhibit the cardiac fibrosis induced by ISO,which is mainly through the blockage of cardiac autoph-agy.
