【目的】探讨DLL4基因在非浸润性膀胱癌中的表达及其与预后的差异。【方法】选择本院2011年5月至2012年10月收治的40例非浸润性膀胱癌部分切除术患者,其中男21例,女19例。以上研究对象均经病理组织学检查证实为膀胱移行细胞癌(BTCC),采用实时荧光定量聚合酶链式反应(RT‐PCR)测定标本中DLL4‐mRNA的表达量,免疫组化检测DLL4蛋白表达情况;对患者术后进行2年随访,分析高表达与肿瘤转移及复发的关系。【结果】DLL4‐mRNA及蛋白在BTCC中呈高表达,癌旁组织中呈低表达,且表达呈现一致性,差异有显著性( P<0.05);同时膀胱癌组织中表达强度存在差异,其中低阳性表达27例,高阳性表达13例;2年随访率为100%,其中高阳性表达患者复发或转移8例,低阳性表达患者复发或转移6例。【结论】DLL4在BTCC的高阳性表达与BTCC复发或转移存在联系;抑制或阻断DLL4信号通路可能抑制BTCC的发生发展。该基因具有一定的临床应用前景。
【目的】探討DLL4基因在非浸潤性膀胱癌中的錶達及其與預後的差異。【方法】選擇本院2011年5月至2012年10月收治的40例非浸潤性膀胱癌部分切除術患者,其中男21例,女19例。以上研究對象均經病理組織學檢查證實為膀胱移行細胞癌(BTCC),採用實時熒光定量聚閤酶鏈式反應(RT‐PCR)測定標本中DLL4‐mRNA的錶達量,免疫組化檢測DLL4蛋白錶達情況;對患者術後進行2年隨訪,分析高錶達與腫瘤轉移及複髮的關繫。【結果】DLL4‐mRNA及蛋白在BTCC中呈高錶達,癌徬組織中呈低錶達,且錶達呈現一緻性,差異有顯著性( P<0.05);同時膀胱癌組織中錶達彊度存在差異,其中低暘性錶達27例,高暘性錶達13例;2年隨訪率為100%,其中高暘性錶達患者複髮或轉移8例,低暘性錶達患者複髮或轉移6例。【結論】DLL4在BTCC的高暘性錶達與BTCC複髮或轉移存在聯繫;抑製或阻斷DLL4信號通路可能抑製BTCC的髮生髮展。該基因具有一定的臨床應用前景。
【목적】탐토DLL4기인재비침윤성방광암중적표체급기여예후적차이。【방법】선택본원2011년5월지2012년10월수치적40례비침윤성방광암부분절제술환자,기중남21례,녀19례。이상연구대상균경병리조직학검사증실위방광이행세포암(BTCC),채용실시형광정량취합매련식반응(RT‐PCR)측정표본중DLL4‐mRNA적표체량,면역조화검측DLL4단백표체정황;대환자술후진행2년수방,분석고표체여종류전이급복발적관계。【결과】DLL4‐mRNA급단백재BTCC중정고표체,암방조직중정저표체,차표체정현일치성,차이유현저성( P<0.05);동시방광암조직중표체강도존재차이,기중저양성표체27례,고양성표체13례;2년수방솔위100%,기중고양성표체환자복발혹전이8례,저양성표체환자복발혹전이6례。【결론】DLL4재BTCC적고양성표체여BTCC복발혹전이존재련계;억제혹조단DLL4신호통로가능억제BTCC적발생발전。해기인구유일정적림상응용전경。
[Objective] To explore the expression and prognostic significance of DLL 4 gene in non‐invasive bladder cancer .[Methods]Surgical specimens were collected from 40 cases of non‐invasive bladder cancer between May 2011 and October 2012 .The definite diagnosis of bladder transitional cell carcinoma (BTCC) was confirmed by histopathological examination .All specimens contained normal bladder tissue around carcinoma (adjacent non‐tumorous bladder tissue ,ANBT ) by 1~2 cm .Reverse transcription‐polymerase chain reaction (RT‐PCR) was employed for detecting the expression of DLL4‐mRNA and immunohistochemistry for the expression of DLL4 pro‐tein .During a follow‐up period of 2 years ,tumor metastasis and recurrence time were observed for analyzing the relationship with DLL4 .[Results] DLL4 mRNA and protein were expressed at high levels in bladder urothelial carcinoma and low levels in adjacent tissues .And the differences were statistically significant ( P <0 .05) .And the expression intensity differed in bladder cancer tissues .The expressions were low ( n=27) and high ( n=13) . The follow‐up rate was 100% .Recurrence or metastasis occurred in high ( n=8) and low ( n=6) expression ca‐ses .[Conclusion] A high expression of DLL4 may play some roles in the recurrence or metastasis of non‐invasive bladder urothelial carcinoma .And blocking the signaling pathways of DLL4 has the potentials of inhibiting the oc‐currence of bladder urothelial carcinoma .
