目的:探讨围绝经期妇女外周血单个核细胞共刺激分子 PD-1mRNA 和 T 细胞表面 PD-1分子的表达和意义。方法采用 Taqman 探针实时荧光定量 PCR 法、流式细胞术检测70例围绝经期、40例绝经后期和30例育龄期组外周血单个核细胞 PD-1mRNA 的表达。结果 PD-1mRNA 的表达在三组间比较差异有统计学意义(P <0.05),与育龄期组相比,围绝经期组及绝经后期组 PD-1mRNA 表达量均显著增高,差异有统计学意义(P <0.05);T 细胞表面 PD-1的表达在三组间比较差异有统计学意义(P <0.05),围绝经期及绝经后期组 CD4+T 细胞和 CD8+T 细胞的 PD-1水平低于育龄期组(P <0.05)。结论共刺激分子 PD-1的异常表达可能与围绝经期妇女免疫功能失调有关。

목적:탐토위절경기부녀외주혈단개핵세포공자격분자 PD-1mRNA 화 T 세포표면 PD-1분자적표체화의의。방법채용 Taqman 탐침실시형광정량 PCR 법、류식세포술검측70례위절경기、40례절경후기화30례육령기조외주혈단개핵세포 PD-1mRNA 적표체。결과 PD-1mRNA 적표체재삼조간비교차이유통계학의의(P <0.05),여육령기조상비,위절경기조급절경후기조 PD-1mRNA 표체량균현저증고,차이유통계학의의(P <0.05);T 세포표면 PD-1적표체재삼조간비교차이유통계학의의(P <0.05),위절경기급절경후기조 CD4+T 세포화 CD8+T 세포적 PD-1수평저우육령기조(P <0.05)。결론공자격분자 PD-1적이상표체가능여위절경기부녀면역공능실조유관。
Objective To investigate the expression of PD-1 mRNA in peripheral blood mononuclear cells and the expression of PD-1 on T cells in perimenopausal and postmenopausal women. Methods The mRNA expression of PD-1 mRNA were detected by Taqman probe real-time quantitative PCR,and the levels of PD-1 on T cells were determined by Flow Cytometry.The subjects in the study included 70 perimenopausal women,40 postmenopausal women and 30 childbearing age women. Results The expression ofPD-1 mRNA differed significantly in these three group(P < 0.05),compared with the childbearing age group,PD-1 mRNA level in postmenopausal women and postmenopausal women was significantly increased(P < 0.05);The expression ofPD-1 on T cells differed significantly in these three group(P < 0.05),compared with the childbearing age group,PD-1 level in postmenopausal women and postmenopausal women was significantly decreased(P < 0.05). Conclusion The abnormal expression of PD-1 may be closely related to the immune dysfunction of perimenopausal women.
