目的:观察辛伐他汀对老年大鼠心肌细胞中炎性因子白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)表达的影响。方法以24月龄大鼠为实验对象,按常规方法分离、剪碎并消化心室肌组织,以含10%胎牛血清的DMEM培养心肌细胞。将培养细胞分为衰老模型对照组、溶剂对照组(DMSO)和辛伐他汀1和10μmol/L组,各组细胞经相应处理后,分别用RT-PCR和Western印迹法检测IL-1β和TNF-α的mRNA及蛋白表达水平,并比较其变化。结果 RT-PCR结果显示,与衰老模型组相比,溶剂组IL-1β和TNF-αmRNA表达无明显变化(P>0.05),辛伐他汀1和10μmol/L组IL-1β和TNF-αmRNA表达均明显降低(分别为P<0.05和P<0.01);与溶剂对照组相比,辛伐他汀1和10μmol/L组IL-1β和TNF-αmRNA表达均明显降低(分别为P<0.05和P<0.01);Western印迹法结果显示,与衰老模型组相比,溶剂对照组IL-1β蛋白表达无明显变化,TNF-α蛋白表达明显降低(P<0.01),辛伐他汀1和10μmol/L组IL-1β和TNF-α蛋白表达均明显降低(P<0.01);与溶剂对照组相比,辛伐他汀1和10μmol/L组IL-1β蛋白表达均明显降低(P<0.01),而TNF-α蛋白表达无明显变化(P>0.05)。结论辛伐他汀可下调衰老大鼠心肌细胞炎性因子IL-1β和TNF-α的基因表达。

목적:관찰신벌타정대노년대서심기세포중염성인자백세포개소-1β(IL-1β)화종류배사인자-α(TNF-α)표체적영향。방법이24월령대서위실험대상,안상규방법분리、전쇄병소화심실기조직,이함10%태우혈청적DMEM배양심기세포。장배양세포분위쇠로모형대조조、용제대조조(DMSO)화신벌타정1화10μmol/L조,각조세포경상응처리후,분별용RT-PCR화Western인적법검측IL-1β화TNF-α적mRNA급단백표체수평,병비교기변화。결과 RT-PCR결과현시,여쇠로모형조상비,용제조IL-1β화TNF-αmRNA표체무명현변화(P>0.05),신벌타정1화10μmol/L조IL-1β화TNF-αmRNA표체균명현강저(분별위P<0.05화P<0.01);여용제대조조상비,신벌타정1화10μmol/L조IL-1β화TNF-αmRNA표체균명현강저(분별위P<0.05화P<0.01);Western인적법결과현시,여쇠로모형조상비,용제대조조IL-1β단백표체무명현변화,TNF-α단백표체명현강저(P<0.01),신벌타정1화10μmol/L조IL-1β화TNF-α단백표체균명현강저(P<0.01);여용제대조조상비,신벌타정1화10μmol/L조IL-1β단백표체균명현강저(P<0.01),이TNF-α단백표체무명현변화(P>0.05)。결론신벌타정가하조쇠로대서심기세포염성인자IL-1β화TNF-α적기인표체。
Objective To investigate the effect of simvastatin on the expression of interleukin-1β(IL-1β) and tumor necrosis factorα(TNF-α) in cardiomyocyte of aging rat. Methods Primary cultures of cardiomyocyte were got from aging rats. Myocyte were divided into control group, dimerthyl sulfoxide(DMSO) group, and simvastatin group, which were treated respectively by cell culture medium, DMSO and simvastatin (1 and 10 μmol/L). The expression of IL-1β、 TNF-α mRNA was evalulated by RT-PCR, and contents of IL-1βand TNF-αprotein were detected by Western blot. Results RT-PCR showed that, compared with the aging model group, IL-1β and TNF-α mRNA expression of solvent group was no significant change (P>0.05), the mRNA expression of IL-1βand TNF-αin 1 and 10μmol/L simvastatin group was significantly lower (P<0.05 and P<0.01); compared with solvent group, in 1 and 10μmol/L simvastatin group IL-1βand TNF-αmRNA expression were significantly lower(P<0.05 and P<0.01). Western-blot results showed that, compared with the aging model group, IL-1β expression of solvent group was no significant change, TNF-α protein expression was significantly lower (P<0.01), in 1 and 10 μmol/L simvastatin group IL-1β and TNF-α protein expression was significantly lower (P<0.01); compared with the solvent group, IL-1β expression in 1 and 10 μmol/L simvastatin group were significantly lower(P<0.01), while TNF-αprotein no significant changed(P>0.05). Conclusions Simvastatin down-regulates IL-1βand TNF-αgene expression.
