目的:观察通心络胶囊对糖尿病周围神经病变小鼠的作用。方法 KK/Upj-Ay小鼠随机分为4组,模型组与高、中、低3个剂量通心络组(0.8,0.4,0.2 g? kg -1),另设C57BL/6小鼠为对照组。灌胃给药12周,测定运动神经传导速度( MNCV)和感觉神经传导速度( SNCV);实时定量荧光聚合酶链反应和蛋白质印迹法测定坐骨神经肿瘤坏死因子-α( TNF-α)、白介素-6(IL-6)、白介素-1β(IL-1β)和核因子-κB(NF-κB)表达。结果与模型组比较,通心络组的小鼠MNCV、SNCV明显增快( P<0.05, P<0.01), TNF-α、IL-6、IL-1β和NF-κB的表达显著下降(P<0.05,P<0.01)。结论通心络胶囊可抑制NF-κB通路,抑制糖尿病周围神经病变小鼠炎症因子表达,从而改善糖尿病周围神经病变。
目的:觀察通心絡膠囊對糖尿病週圍神經病變小鼠的作用。方法 KK/Upj-Ay小鼠隨機分為4組,模型組與高、中、低3箇劑量通心絡組(0.8,0.4,0.2 g? kg -1),另設C57BL/6小鼠為對照組。灌胃給藥12週,測定運動神經傳導速度( MNCV)和感覺神經傳導速度( SNCV);實時定量熒光聚閤酶鏈反應和蛋白質印跡法測定坐骨神經腫瘤壞死因子-α( TNF-α)、白介素-6(IL-6)、白介素-1β(IL-1β)和覈因子-κB(NF-κB)錶達。結果與模型組比較,通心絡組的小鼠MNCV、SNCV明顯增快( P<0.05, P<0.01), TNF-α、IL-6、IL-1β和NF-κB的錶達顯著下降(P<0.05,P<0.01)。結論通心絡膠囊可抑製NF-κB通路,抑製糖尿病週圍神經病變小鼠炎癥因子錶達,從而改善糖尿病週圍神經病變。
목적:관찰통심락효낭대당뇨병주위신경병변소서적작용。방법 KK/Upj-Ay소서수궤분위4조,모형조여고、중、저3개제량통심락조(0.8,0.4,0.2 g? kg -1),령설C57BL/6소서위대조조。관위급약12주,측정운동신경전도속도( MNCV)화감각신경전도속도( SNCV);실시정량형광취합매련반응화단백질인적법측정좌골신경종류배사인자-α( TNF-α)、백개소-6(IL-6)、백개소-1β(IL-1β)화핵인자-κB(NF-κB)표체。결과여모형조비교,통심락조적소서MNCV、SNCV명현증쾌( P<0.05, P<0.01), TNF-α、IL-6、IL-1β화NF-κB적표체현저하강(P<0.05,P<0.01)。결론통심락효낭가억제NF-κB통로,억제당뇨병주위신경병변소서염증인자표체,종이개선당뇨병주위신경병변。
Objective To investigate the effect of Tongxinluo capsule on diabetic peripheral neuropathy in mice .Methods KK/Upj -Ay mice were randomly divided into 4 groups,model, Tongxinluo low, middle, high doses groups (0.8, 0.4, 0.2 g? kg -1 ).C57BL/6 mice were se-lected as control group.Mice were given intragastrically for 12 weeks. Motor nerve conduction velocity ( MNCV ) and sensory nerve conduction velocity ( SNCV) were detected.The expression of tumor necrosis factor alpha ( TNF -α) , interleukin 6 ( IL -6 ) and interleukin 1 beta (IL-1β) and nuclear factor kappa B (NF-κB) in sciatic nerve were examined by real time PCR and Western blot .Results Compared with model group , MNCV and SNCV were faster ( P<0.05 , P<0.01 ) .The TNF-α, IL-6 , IL-1βand NF-κB expression significantly were de-creased in Tongxinluo group ( P < 0.05 , P < 0.01 ) . Conclusion Tongxinluo capsule inhibits inflammatory cytokines expres-sion in diabetic peripheral neuropathy mice by inhibiting NF -κB pathway , to ameliorates diabetic peripheral neuropathy .
