目的::研究2型糖尿病大鼠肝脏胰岛素受体(insulin receptor,IR)表达的变化及丝胶的保护作用。方法:36只SD大鼠随机分为正常对照组、糖尿病模型组和丝胶治疗组3组,每组12只大鼠。采用链脲佐菌素连续腹腔注射法制作2型糖尿病大鼠模型,待模型成功建立后,丝胶治疗组大鼠给予丝胶(2.4g/kg/d)灌胃35d。采用葡萄糖氧化酶法检测各组大鼠的空腹血糖,Western Blotting法和RT-PCR检测大鼠肝脏IR蛋白和mRNA的表达。结果:与糖尿病模型组大鼠比较,丝胶治疗组大鼠肝脏IR蛋白和mRNA的表达明显升高(P<0.01)。结论:丝胶可能通过上调2型糖尿病大鼠肝脏IR的表达在降血糖中发挥作用。

목적::연구2형당뇨병대서간장이도소수체(insulin receptor,IR)표체적변화급사효적보호작용。방법:36지SD대서수궤분위정상대조조、당뇨병모형조화사효치료조3조,매조12지대서。채용련뇨좌균소련속복강주사법제작2형당뇨병대서모형,대모형성공건립후,사효치료조대서급여사효(2.4g/kg/d)관위35d。채용포도당양화매법검측각조대서적공복혈당,Western Blotting법화RT-PCR검측대서간장IR단백화mRNA적표체。결과:여당뇨병모형조대서비교,사효치료조대서간장IR단백화mRNA적표체명현승고(P<0.01)。결론:사효가능통과상조2형당뇨병대서간장IR적표체재강혈당중발휘작용。
Objective:To study the changes of insulin receptor (IR) expression in liver of type 2 diabetes mellitus rats and protective actions of sericin.Methods:36 male SD rats were randomly divided into the following 3 groups (n=12):normal control group, diabetes model group and sericin treatment group. The rats with type 2 diabetes mellitus were induced by continuous intraperitoneal injection of streptozotocin. After successfully establishing diabetes rats model, the rats in sericin treatment group were lavaged with sericin (2.4g/kg/d) for 35 days. Glucose oxidase was used to detect the fasting blood glucose; Western Blotting and RT-PCR were used to detect the IR protein and mRNA expression in the liver of rats.Results:Compared with rats in diabetes model group, the IR protein and mRNA expression in liver of rats in sericin treatment group increased obviously (P<0.01).Conclusions: One of the mechanisms of sericin decreasing blood glucose maybe secrin can upregulate IR expression in liver of type 2 diabetes mellitus rats.
