目的:探讨短时低速离心法在酶联免疫吸附测定(ELISA)中代替传统拍板的可行性。方法:采用间接ELISA,7例样本设置副孔,一组采用短时低速离心法,一组采用传统手拍板,传统手拍组两个96孔板,短时低速离心组四个96孔板,每板样本相同。结果:采用短时低速离心可以在相同时间内完成四个96孔板操作,而传统手拍板只能完成两块96孔板操作。通过独立样本t检验分析比较两组吸光度值,短时低速离心组同一样本最终检测均值与传统手拍板组之间差异无统计学意义(P>0.05),两种方法检测效果相差不大,但最终检测值稳定性比传统手拍板组更高。结论:采用短时低速离心法更为简单易行,操作稳定,对于间接ELISA检测大批量样本来说,可节省至少一倍时间。

ObjectiveDiscusses the short low speed centrifugation in enzyme linked immunosorbent assay (ELISA) instead of the traditional clappers feasibility.Methods By indirect ELISA, 7 cases of sample was set vice hole , a set of uses short low speed centrifugation, a set of uses the traditional hand clappers. Traditional hand clappers on group of two 96 - well plates, short-term low speed centrifuge group four 96 - well plates, each plate samples are the same.Results Using short low speed centrifuge can be finished four pieces 96 - well plates at the same time , traditional hand clappers can complete two pieces of 96 - well plates. Through independent sample t-test analysis, compare two groups of absorbance value, short-term low speed centrifuge group the stability of the final values higher than the traditional hand clappers group.Conclusions Use short low speed centrifuge method is more simple, the operation stability, for indirect ELISA to detect large quantities of samples, can save at least one half time.
