目的:探讨雷公藤内酯醇对糖尿病大鼠心肌内葡萄糖转运体(Glut)-1、Glut-4表达的干预。方法雄性SD大鼠60只,按体质量随机分为正常对照组10只和糖尿病模型组50只。糖尿病模型组一次性腹腔注射链脲佐菌素53 mg/kg,正常对照组仅腹腔注射等体积0.1 mol/L柠檬酸缓冲液。注射链脲佐菌素72 h后,尾静脉采血测空腹血糖并同时测尿糖,血糖≥16.7 mmol/L、尿糖(+++)以上者列入糖尿病观察对象。将造模成功的48只雄性大鼠,按体质量、空腹血糖随机分为糖尿病模型对照组(12只),雷公藤内酯醇小、中、大剂量组(各12只)。结果雷公藤内酯醇中、大剂量组大鼠心体比值均比糖尿病对照组降低,但都高于正常对照组;雷公藤内酯醇中、大剂量组大鼠心肌中Glut-1、Glut-4的表达均比糖尿病对照组有显著增高,但都较正常对照组明显减少;雷公藤内酯醇小剂量组糖尿病大鼠心体比值、心肌中Glut-1、Glut-4的表达与糖尿病对照组相比变化不明显。结论雷公藤内酯醇能明显上调糖尿病大鼠心肌细胞内Glut-1、Glut-4的表达,改善高血糖对糖尿病大鼠心肌的毒性作用。

목적:탐토뢰공등내지순대당뇨병대서심기내포도당전운체(Glut)-1、Glut-4표체적간예。방법웅성SD대서60지,안체질량수궤분위정상대조조10지화당뇨병모형조50지。당뇨병모형조일차성복강주사련뇨좌균소53 mg/kg,정상대조조부복강주사등체적0.1 mol/L저몽산완충액。주사련뇨좌균소72 h후,미정맥채혈측공복혈당병동시측뇨당,혈당≥16.7 mmol/L、뇨당(+++)이상자렬입당뇨병관찰대상。장조모성공적48지웅성대서,안체질량、공복혈당수궤분위당뇨병모형대조조(12지),뢰공등내지순소、중、대제량조(각12지)。결과뢰공등내지순중、대제량조대서심체비치균비당뇨병대조조강저,단도고우정상대조조;뢰공등내지순중、대제량조대서심기중Glut-1、Glut-4적표체균비당뇨병대조조유현저증고,단도교정상대조조명현감소;뢰공등내지순소제량조당뇨병대서심체비치、심기중Glut-1、Glut-4적표체여당뇨병대조조상비변화불명현。결론뢰공등내지순능명현상조당뇨병대서심기세포내Glut-1、Glut-4적표체,개선고혈당대당뇨병대서심기적독성작용。
Objective To discuss intervention of triptolide on glucose transporter type (Glut)-1, Glut-4 expres-sion in the myocardium of diabetic rats. Methods Sixty SD rats were randomly divided into normal control group (n=10) and diabetes model (DM)group (n=50). Diabetic model group were injected of streptozotocin (STZ) 53 mg/kg intraperitoneally, the normal control group were injected volume of the buffer. After the injection of STZ 72 h , fasting glucose and simultaneously measuring urine were tested from tail vein blood , both blood glucose≥16.7 mmol/L and rather than urine (+++) were included in the DM observed object. The successful model of 48 male rats , according to body weight, fasting blood glucose, DM model were randomly divided into a control group (12), triptolide (TP) of small, medium and large-dose group (12 each). Results TP in medium (1.8 g/kg) and high dose (2.2 g/kg) of rats heart body ratio were lower than DM group significantly, but were higher than the normal control group; the expression of TP in medium and high dosage groups in myocardium of rats with Glut-1 and Glut-4 were significantly higher than DM group. But all decreased significantly comparing with normal group; TP group of small dose of diabetic rat heart body ratio, myocardial Glut-1, Glut-4 expression comparing with the DM group did not change significantly. Conclusion Triptolide significantly increases Glut-1 and Glut-4 expression in diabetic rat myocardial cells, improves the toxic effect of hyper-glycemia on myocardium of DM rats.
