目的：探讨黄芩苷对人胃癌 SGC-7901细胞增殖抑制、诱导凋亡作用及其分子作用机制。方法 MTT 法检测黄芩苷对 SGC-7901细胞的增殖抑制作用；PI 单染法检测细胞周期；Annexin V/PI 双染法检测细胞凋亡；Western blot 法检测凋亡相关基因 Bcl-2和 Bax 的蛋白表达水平。结果黄芩苷对 SGC-7901细胞生长有明显抑制作用，作用24 h 和48 h 对 SGC-7901细胞的半数抑制浓度（IC50）分别为218.73μmol·L-1，161.86μmol·L-1，呈现时间—剂量依赖性；FCM 结果显示黄芩苷能使 SGC-7901细胞阻滞在 G2/M 期；Annexin V/PI 双染法验证黄芩苷可以诱导细胞凋亡；Western blot 结果显示 Bax 蛋白水平明显上调，而 Bcl-2的蛋白表达水平降低。结论黄芩苷对 SGC-7901细胞增殖具有显著的抑制作用并可诱导细胞凋亡，其作用机制与其上调 Bax 蛋白表达水平的同时，下调 Bcl-2蛋白表达水平，从而诱导细胞凋亡有关。
목적：탐토황금감대인위암 SGC-7901세포증식억제、유도조망작용급기분자작용궤제。방법 MTT 법검측황금감대 SGC-7901세포적증식억제작용；PI 단염법검측세포주기；Annexin V/PI 쌍염법검측세포조망；Western blot 법검측조망상관기인 Bcl-2화 Bax 적단백표체수평。결과황금감대 SGC-7901세포생장유명현억제작용，작용24 h 화48 h 대 SGC-7901세포적반수억제농도（IC50）분별위218.73μmol·L-1，161.86μmol·L-1，정현시간—제량의뢰성；FCM 결과현시황금감능사 SGC-7901세포조체재 G2/M 기；Annexin V/PI 쌍염법험증황금감가이유도세포조망；Western blot 결과현시 Bax 단백수평명현상조，이 Bcl-2적단백표체수평강저。결론황금감대 SGC-7901세포증식구유현저적억제작용병가유도세포조망，기작용궤제여기상조 Bax 단백표체수평적동시，하조 Bcl-2단백표체수평，종이유도세포조망유관。
Objective To investigate the inhibiting effect and its mechanism of baicalin on human gastric carcinoma SGC-7901 cells. Methods Growth inhibition rates of SGC-7901 cells were measured with MTT method, the cell cycle distribution was detected by PI single staining, apoptosis was detected by Annexin V/PI double staining, and the expression levels of Bcl-2, bax protein were analyzed by western blot. Results Baicalin inhibited the proliferation of SGC-7901 cells in a dose and time dependent manner. The IC50 of 24 h and 48 h were 218.73 μmol·L-1 and 161.86 μmol·L-1 respectively. The cell growth was arrested in G2/M phase. Fluorescent staining can detect the apoptotic cells induced by baicalin. The expression of Bax protein was increased while the Bcl-2 was decreased. Conclusion Baicalin not only inhibits proliferation but also induces apoptosis of SGC-7901 cells. Its mechanism is related to down-regulating Bcl-2 while up-regulating Bax.
