脑血管疾病是一种严重威胁人类特别是50岁以上中老年人生命和健康的常见病。因脑血管疾病死亡的病例中约1/4死于颅内动脉瘤( cerebral aneurysm ,CA)破裂。因此,有效降低CA的发病率并减少其并发症的危害对于维护人类健康、延长人类平均寿命具有非常重要的意义。 CA血管壁组织中的蛋白水解酶活性较正常动脉管壁组织明显增强,纤维蛋白溶解酶、膜型基质金属蛋白酶-1、基质金属蛋白酶( matrix metallo-proteinase ,MMP )-2的表达水平也明显升高。而MMP的抑制剂如多西环素、他汀类药物可以抑制CA的形成和发展。 MMP参与血管重塑的多个病理环节,如形态重构、血管生成、凋亡和组织修复等。作者对国内外相关文献分析,阐述MMP及其抑制剂在颅内动脉瘤形成和发展过程中的生物学作用,为广大医生提供参考。

Cerebral diseases is one of the commonest health hazards that severely threaten hu-man′s health, especially the aged more than 50 yrs old.And about one-fourth of cerebrovascular deaths are due to ruptured cerebral aneurysms (CA).Therefore, exploring effective methods to de-crease CA′s incidence and reduce the effects of complications is of vital importance for maintaining-people′s health and extending the average human life span .It has been observed that the proteolytic activity , the expression of plasmin , membranetype-1 matrix metalloproteinase-1, and matrix metallo-proteinase-2 ( MMP-2) increased in aneurysm wall tissue that were all more than those in normal cerebral arteries .Moreover , MMP inhibitors such as doxycycline and statins may prohibit aneurysm formation and growth .MMP is involved in a lot of pathophysiology processes such as morphogenesis , angiogenesis , apoptosis and tissue repair .In this article , we reviewed the latest literatures and ex-plained in detail the roles of MMP and its inhibitors in the formation and growth process of CA .
