目的:探讨葡萄糖浓度波动对单核细胞表达CD11b及肿瘤坏死因子-α( TNF-α)的影响。方法根据葡萄糖浓度将THP-1细胞株随机分为对照组、高葡萄糖组、葡萄糖浓度波动组和渗透压控制组。采用流式细胞仪检测单核细胞表面CD11b表达,酶联免疫法测定TNF-α浓度,并进行各组间的比较。结果高葡萄糖组CD11b、TNF-α浓度高于对照组,葡萄糖浓度波动组高于高葡萄糖组,渗透压控制组介于高葡萄糖组和葡萄糖浓度波动组之间(P均<0.05)。结论葡萄糖浓度波动较单纯高糖更能促进单核细胞分泌CD11b、TNF-α。

목적:탐토포도당농도파동대단핵세포표체CD11b급종류배사인자-α( TNF-α)적영향。방법근거포도당농도장THP-1세포주수궤분위대조조、고포도당조、포도당농도파동조화삼투압공제조。채용류식세포의검측단핵세포표면CD11b표체,매련면역법측정TNF-α농도,병진행각조간적비교。결과고포도당조CD11b、TNF-α농도고우대조조,포도당농도파동조고우고포도당조,삼투압공제조개우고포도당조화포도당농도파동조지간(P균<0.05)。결론포도당농도파동교단순고당경능촉진단핵세포분비CD11b、TNF-α。
Objective To investigate the effectss of fluctuation of glucose concentrations on CD 11b and tumor necrosis factor ( TNF)-αproduced by Mmonocytes , and discuss the role of the blood glucose in the pathogenesis of diabetic mac-roangiopathy .Methods The THP-1 cell strain was randomly divided into the control group , high glucose group , fluctua-ting glucose concentration group and osmotic pressure control group .The expression of CD11b on the surface of monocytes was detected by flow cytometry .The concentrations of TNF-αwere detected by ELISA and then were compared in each group.Results The concentrations of TNF-αand CD11b in the high glucose group were significantly higher than those in the control group, the levels of TNF-αand CD11b of the fluctuating glucose group were higher than those of the high glu-cose group, and the concentrations of TNF-αand CD11b of the osmotic pressure control group were between those of the high glucose group and the fluctuating glucose group .The differences between groups was statistically significant ( all P<0.05).Conclusion The fluctuation of glucose can promote monocytes secreting more CD 11b and TNF-α, which can pro-duce a greater cellular toxicity than simple high glucose does .
